榄香烯乳联合化疗对Lewis肺癌的影响及机制

摘　　要：目的探讨榄香烯乳联合化疗对Lewis肺癌的影响及机制。方法选取近交系C57BL/6小鼠40只,所有小鼠建立Lewis肺癌小鼠模型,随机分为模型组、榄香烯乳组、化疗组和联合组。榄香烯乳组腹腔注射100 mg/kg/d榄香烯乳,化疗组腹腔注射3 mg/kg/d足叶乙甙和3 mg/kg/d顺铂,联合组腹腔注射100 mg/kg/d榄香烯乳、3 mg/kg/d足叶乙甙和3 mg/kg/d顺铂,连续7 d,观察各组瘤体质量、脾脏和胸腺指数,检测血管内皮生长因子(VEGF),Western blot检测Per2和SIRT1蛋白表达。结果榄香烯乳组、化疗组和联合组瘤重和肺转移结节数均较模型组降低(P<0.05),其中联合组瘤重和肺转移结节数分别为(2.64±0.78)g和(1.12±0.35)个,明显低于榄香烯乳组、化疗组(P<0.05)。联合组脾脏指数和胸腺指数分别为(3.00±0.60)mg/g和(2.03±0.36)mg/g,明显高于化疗组和模型组(P<0.05),但低于榄香烯乳组(P<0.05)。榄香烯乳组、化疗组和联合组血清VEGF均较模型组降低(P<0.05),其中联合组血清VEGF为(66.72±19.12)pg/ml,明显低于榄香烯乳组、化疗组(P<0.05);联合组肿瘤组织Per2和SIRT1蛋白相对表达量分别为(0.410±0.100)和(0.722±0.106),明显高于榄香烯乳组、化疗组和模型组(P<0.05)。结论榄香烯乳联合化疗对Lewis肺癌有较好的抑瘤作用,可能与免疫调节、VEGF下调,促进Per2/SIRT1蛋白表达有关。Objective To investigate the effect and mechanism of elemene emulsion combined with chemotherapy on Lewis lung cancer.Methods 40 inbred C57BL/6 mice were selected to establish Lewis lung cancer model,all mice were randomly divided into model group,Elemene Emulsion group,chemotherapy group and combination group,the Elemene Emulsion group was intraperitoneally injected with 100 mg/kg/d elemene emulsion,the chemotherapy group was intraperitoneally injected with 3 mg/kg/d etoposide and 3 mg/kg/d cisplatin,and the combination group was intraperitoneally injected with 100 mg/kg/d elemene emulsion,mg/kg/d etoposide and 3 mg/kg/d cisplatin for 7 consecutive days,tumor body mass,spleen and thymus index were observed,vascular endothelial growth factor(VEGF)was detected,and Per2 and SIRT1 protein expression were detected by Western blot.Results The tumor weight and the number of pulmonary metastatic nodules in Elemene Emulsion group,chemotherapy group and combination group were all lower than those in model group(P<0.05),the tumor weight and the number of pulmonary metastatic nodules in combination group were(2.64±0.78)g and(1.12±0.35),which were significantly lower than those in Elemene Emulsion group and chemotherapy group(P<0.05);The spleen index and thymus index of the combined group were(3.00±0.60)mg/g and(2.03±0.36)mg/g,which were significantly higher than those of the chemotherapy group and the model group(P<0.05),but lower than those of the Elemene Emulsion group(P<0.05);The serum levels of VEGF in Elemene Emulsion group,chemotherapy group and combination group were lower than those in model group(P<0.05),the serum levels of VEGF in combination group were(66.72±19.12)pg/ml,which were significantly lower than those in Elemene Emulsion group and chemotherapy group(P<0.05);The relative expression levels of Per2 and SIRT1 protein in the combined group were(0.410±0.100)and(0.722±0.106),which were significantly higher than those in the Elemene Emulsion group,the chemotherapy group and the model group(P<0.05).Conclu
