摘 要:MyoG基因是对与产肉性状相关的重要候选基因之一。筛选MyoG基因与胴体体尺性状相关的遗传变异分子标记,对开展肉用生产性状分子选育具有重要的意义。本研究以特克塞尔×哈萨克横交F2代绵羊为实验群体,进行MyoG基因的SNPs多态性与胴体体尺性状之间的相关性分析。首先,随机选择50只特哈横交F2代绵羊,采用DNA双向测序技术筛选MyoG基因单核苷酸多态位点(SNPs),得到6个SNP位点。然后,采用Sequenom MassARRAY○RSNP检测方法对384只绵羊MyoG基因6个SNPs进行分型。并将得到434只绵羊的6个SNPs基因型与其胴体重、胴体长、胸宽、肩宽、胸深、臀宽和踝骨宽7个胴体性状进行关联分析结果显示MyoG基因rs407552631、rs40016030、rs600781279和rs4102122554个SNPs与胴体体尺性状相关,它们均位于内含子区域。其中,rs600781279中的GG和AG与个体踝骨宽显著相关;rs407552631中的GT和TT与胴体长、肩宽、臀宽和踝骨宽显著相关;rs410212255中的GA和GG与胸深显著相关;rs400160301中的GA与胴体长、臀宽和踝骨宽显著相关,AA与胴体长、肩宽、胸深、臀宽和踝骨宽显著相关,GG与肩宽、胸深和臀宽显著相关。本研究为今后绵羊胴体性状分子选育提供了参考。MyoG gene is one of the important candidate genes related to meat production traits.Screening of genetic variant molecular markers of MyoG gene related to carcass size traits is of great significance to the development of molecular breeding for meat production traits.In this study,F2 generation crossed by Texel and Kazakh sheep were used as the experimental animals to analyze the correlation between the SNPs polymorphism of the MyoG gene and the carcass size traits.First,50 F2 generation sheep were randomly selected,and single nucleotide polymorphisms(SNPs)of MyoG gene were screened by DNA two-way sequencing technology,and 6 SNPs were obtained.Then,Sequenom MassARRAY RSNP detection method was used to carry out the typing of 6 SNPs of MyoG gene in 384 sheep.And the genotypes of 6 SNPs were associated with 7 carcass traits consisting of carcass weight,carcass length,chest width,shoulder width,chest depth,hip width and ankle width.The results showed that the 4 SNPs of rs407552631,rs40016030,rs600781279 and rs410212255 of MyoG gene were related to carcass traits,and they were all located in the intron region.Among them,GG and AG in rs600781279 were significantly related to the ankle width of an individual;GT and TT in rs407552631 were significantly related to carcass length,shoulder width,hip width and ankle width;GA and GG in rs410212255 were significantly related to chest depth;GA in rs400160301 was significantly correlated with carcass length,hip width and ankle width,AA was significantly correlated with carcass length,shoulder width,chest depth,hip width and ankle width,and GG was significantly correlated with shoulder width,chest depth and hip width.A reference was provided for future molecular selection and breeding of sheep carcass traits.
