摘  要:目的探讨利用四氯化碳(CCl_(4))联合橄榄油诱导建立家兔肝硬化模型,为临床研究提供可靠的动物模型。方法体重1.8~2.3kg家兔40只,随机分为两组,实验组(n=30)每周腹腔注射2次浓度为50%CCl_(4)橄榄油溶液,剂量分别在实验的第3周、第6周、第9周进行调整。对照组(n=10)腹腔注射等量的生理盐水。实验组和对照组均于注药后第3周、第6周、第9周分别留取家兔的肝脏组织和血清标本进行病理观察和生化检测。结果对照组家兔在实验期间无死亡。实验组在整个实验期间死亡8只,其中前3周死亡5只,~6周死亡2只,~9周死亡1只,,存活率73%.实验组家兔在第9周均形成肝硬化;实验组较对照组第3周、第9周的谷草转氨酶(AST)、谷丙转氨酶(ALT)明显升高,差异有统计学意义(P<0.05)。结论CCl_(4)联合橄榄油可有效建立家兔肝硬化模型。Objective To explore the model of rabbit cirrhosis induced by carbon tetrachloride(CCl_(4))combined olive oil to provide a reliable animal model for clinical studies.Methods Forty rabbits(1.8~2.3kg of weight)were randomly divided into two groups.The experimental group(n=30)received 50%CCl_(4) olive oil solution twice a week,and the dose was adjusted at weeks 3,6 and 9.The control group(n=10)were intraperitoneally injected equal amounts of saline.Liver tissue and serum samples from rabbits were retained at weeks 3,6 and 9,and pathological observation and biochemical tests were performed.Results No rabbits of controls died during the experiment.The experimental group had 8 deaths throughout the entire period,5 died at the first 3 weeks,2 died at 3~6 weeks,and 1 died at 6~9 weeks.The survival rate was 73%.All surviving rabbits in the experimental group developed cirrhosis at week 9;Aspartate aminotransferase and alanine aminotransferase in the experimental group were significantly higher than controls at weeks 3 and 9(P<0.05).Conclusion CCl_(4) and olive oil can effectively establish model of rabbit cirrhosis.
