摘 要:目的探讨叶黄素对急性酒精中毒引起的小鼠胃黏膜损伤的保护作用。方法90只昆明种小鼠随机分为6组:正常对照组,酒精模型组,海王金樽阳性对照组(20 mg/kg),叶黄素低、中、高剂量组(20、40、80 mg/kg),每组15只。阳性对照和叶黄素各组于饮酒前30 min灌胃给药。除正常对照组外,各组使用56°白酒0.02 mL/(g·bw)灌胃。记录小鼠醉酒率和醉酒潜伏期,光学显微镜下观察胃黏膜溃疡情况,HE染色观察小鼠胃组织的病理变化,检测血清中ADH含量和胃组织中SOD、MDA、GSH-Px含量。结果与正常组比较,酒精模型组小鼠ADH含量显著降低,胃黏膜组织出现明显水肿、局部充血、溃疡等症状,MDA含量增高,SOD和GSH-Px活性降低。和酒精模型组对比,叶黄素中、高剂量组小鼠醉酒率显著降低,缩短醉酒时间;HE染色中性粒细胞及淋巴细胞浸润明显减少,水肿程度减轻;MDA含量降低,ADH、SOD和GSH-Px活性明显增高。结论叶黄素能够减少急性酒精中毒对胃黏膜组织损伤,具有预防保护作用。Objective To investigate the protective effect of lutein on gastric mucosal injury induced by acute alcoholism in mice.Methods Ninety Kunming mice were randomly divided into 6 groupsnormal control group,alcohol model group,Haiwang Jinzun positive control group(20 mg/kg),lutein low,medium and high dose groups(20,40 and 80 mg/kg),with 15 mice in each group.The positive control and lutein groups were administered by gavage 30 min before drinking.In addition to normal control group,each group was given 56 degree Baijiu 0.02 mL/(g·bw)gavage.The intoxication rate and intoxication latency of mice were recorded.The gastric mucosal ulcer was observed under optical microscope.The pathological changes of gastric tissue were observed by HE staining.The contents of ADH in serum and SOD,MDA and GSH Px in gastric tissue were detected.Results Compared with the normal group,the content of ADH in the alcohol model group decreased significantly,the symptoms of gastric mucosal tissue such as edema,local congestion and ulcer occurred,the content of MDA increased,and the activities of SOD and GSH Px decreased.Compared with the alcohol model group,the drunkenness rate of mice in medium and high dose lutein groups was significantly reduced and the drunkenness time was shortened;The infiltration of neutrophils and lymphocytes was significantly reduced and the degree of edema was reduced by HE staining;The content of MDA decreased,and the activities of ADH,SOD and GSH Px increased significantly.Conclusion Lutein can reduce the injury of gastric mucosa caused by acute alcoholism and has a preventive and protective effect.
