AMPK、YAP1与E-cadherin蛋白在胃癌组织中的表达及临床意义

摘　　要：目的探讨腺苷酸活化蛋白激酶(adenosine monophosphate activated protein kinase,AMPK)、Yes相关蛋白1(yes-associated protein 1,YAP1)、E-cadherin蛋白在胃癌组织中的表达与临床各病理特征的关系。方法选取2019年1月至2020年1月在宁夏医科大学总医院行胃癌根治术患者的组织标本60例,采用免疫组化法检测AMPK、YAP1、E-cadherin在胃癌组织和相应癌旁正常组织中的表达,统计各基因在胃癌组织中的表达阳性率,分析其与临床各病理特征的关系;进一步分析胃癌组织中AMPK、YAP1、E-cadherin蛋白表达的相关性。结果60例胃癌组织中,AMPK、YAP1、E-cadherin的蛋白表达阳性率分别为40.00%、80.00%、36.67%,而癌旁正常组织中的表达阳性率分别为85.00%、28.33%、75.00%;AMPK、E-cadherin在胃癌组织中的表达阳性率均低于癌旁组织(P均<0.05),YAP1在胃癌组织中的表达阳性率高于癌旁组织(P<0.05)。AMPK的表达与脉管浸润、肿瘤分化程度、淋巴结转移及TNM分期相关(P均<0.05),而与性别、年龄、肿瘤大小无关(P均>0.05);YAP1的表达与肿瘤大小、脉管浸润、肿瘤分化程度、淋巴结转移及TNM分期相关(P均<0.05),而与性别、年龄无关(P均<0.05);E-cadherin的表达与脉管浸润、淋巴结转移及TNM分期相关(P均<0.05),与性别、年龄、肿瘤大小、肿瘤分化程度无关(P均>0.05)。AMPK与YAP1的表达呈负相关(P<0.01),与E-cadherin的表达呈正相关(P<0.01),YAP1与E-cadherin的表达呈负相关(P<0.01)。结论AMPK、YAP1、E-cadherin蛋白均在胃癌组织中异常表达,与胃癌的进展有一定相关性,联合检测可能对胃癌的诊治具有指导价值。Objective To explore the expression of AMPK、YAP1 and E-cadherin proteins in gastric carcinoma and their relationship with clinical pathological characteristics.Methods A total of 60 tissue specimens were collected from patients who underwent radical gastrectomy in the Department of Gastrointestinal Surgery and Department of Oncology of General Hospital of Ningxia Medical University from January 2019 to January 2020.The expression of AMPK,YAP1 and E-cadherin in gastric carcinoma tissues and corresponding adjacent normal tissues were detected by immunohistochemical method.The positive expression rate of each gene in gastric carcinoma tissues was statistically analyzed,and the relationship between the expression and clinical pathological features was analyzed,and the relationship between AMPK,YAP1,and E-cadherin protein expression was further analyzed in gastric cancer tissues.Results In 60 cases of gastric carcinoma tissues,the positive rates of AMPK,YAP1,and E-cadherin protein expression were 40.00%,80.00%,and 36.67%,respectively;in 60 cases of adjacent normal tissues,the positive rates were 85.00%,28.33%,and 75.00%;The positive rates of AMPK and E-cadherin expression in gastric carcinoma tissues were significantly lower than those in adjacent tissues(P all<0.05),and the positive rate of YAP1 expression in gastric carcinoma tissues was significantly higher than that in adjacent tissues(P<0.05).The expression of AMPK was significantly correlated with vascular invasion,tumor differentiation,lymph node metastasis,and TNM stage(P all<0.05),but has no correlation with gender,age,and tumor size(P all>0.05);The expression of YAP1 was significantly correlated with tumor size,vascular invasion,tumor differentiation,lymph node metastasis and TNM stage(P<0.05),but not with gender and age;The expression of E-cadherin was correlated with vascular invasion,lymph node metastasis and TNM stage(P all<0.05),but not correlated with gender,age,tumor size,tumor differentiation,lymph node metastasis,and TNM stage(P all>0.05).The expr
