摘 要:目的建立安徽怀远地区石榴皮中没食子酸(GA)、鞣花酸成分(EA)的含量同时检测方法。方法采用反相高效液相色谱法(RP-HPLC),Symmetry C18(250 mm×4.6 mm,5μm)色谱柱,以甲醇(A)-0.08%磷酸溶液(B)为流动相梯度洗脱,流速1.0 mL/min,柱温30℃,波长270 nm同时检测GA和EA含量。采用SPSS 17.0对测定结果进行分析,比较不同品种石榴皮中各成分的含量差异。结果没食子酸和鞣花酸分别在6.6375~212.4000,6.6625~213.2000μg/mL呈线性关系,平均加样回收率为96.90%(RSD=0.78%,n=6)和97.90(RSD=0.63%,n=6)。结论此方法准确稳定、灵敏方便、重现性好,可作为安徽本地石榴皮部位中GA、EA成分含量的同时检测;且白玉石榴皮中GA、EA含量较红酸石榴皮的含量偏高。Objective To establish a method for simultaneous determination of gallic acid and ellagic acid punicalagin in pomegranate peel in Huaiyuan,Anhui.Method The reversed-phase high performance liquid chromatography was used,with Symmetry C18(250 mm×4.6 mm,5μm)chromatographic column,methanol(A)-0.08%phosphoric acid solution(B)as mobile phase gradient elution,flow rate 1.0 mL/min,Column temperature 30℃,detection wavelength 270 nm to detect gallic acid and ellagic acid content.SPSS 17.0 was used to analyze the measurement results and the differences in the contents of various components in different varieties of pomegranate peel were compared.Results The gallic acid and ellagic acid showed a linear relationship between 6.6375~212.4 and 6.6625~213.2μg/mL,respectively,and the average recovery rate was 96.90(RSD=0.78%,n=6)and 97.90(RSD=0.63%,n=6).Conclusion This method was accurate,stable,sensitive,convenient,and reproducible.It could be used as a simultaneous detection of the content of gallic acid and ellagic acid in the local pomegranate skin of Anhui,and the content of gallic acid and ellagic acid in the white jade pomegranate skin was higher than that of red acid.The content of pomegranate peel was high.
