摘  要:目的利用网络药理学方法研究沙棘油抗炎的作用机制。方法采用气相色谱-质谱联用(GC-MS)法检测沙棘油样品中脂肪酸的组成;通过中药系统药理数据库与分析平台(TCMSP)搜索,Swiss TargetPrediction数据库预测沙棘油有效成分对应靶点;通过DisGeNET等数据库,获取炎症相关靶点;将活性成分和潜在作用靶点导入Cytoscape软件构建沙棘油抗炎的"活性成分-作用靶点"网络。将潜在作用靶点录入STRING平台构建靶点间的蛋白相互作用(PPI)网络。采用DAVID数据库对潜在作用靶点进行GO富集分析和KEGG通路富集分析。结果通过测定并筛选得到肉豆蔻酸、棕榈酸、棕榈油酸等8种活性成分,检索并预测得到208个对应靶点。利用在线网站Venny 2.1.0(https://bioinfogp.cnb.csic.es/tools/venny/index.html)筛选出PPARG、PPARA、PTPN1、PTGES、RORC等19个成分和疾病的交集靶点,涉及信号通路有癌症通路、非酒精性脂肪肝(NAFLD)等。结论本研究发现沙棘油通过多成分多靶点对炎症反应进行调控,为后续研究提供了指导意义。Objective To explore the anti-inflammatory mechanism of sea buckthorn oil by means of network pharmacology.Methods Detect the composition of fatty acids in sea buckthorn oil by gas chromatography-mass spectrometry(GC-MS).The active components and action targets of sea buckthorn oil were retrieved and collected from TCMSP and Swiss TargetPrediction Database.Obtain inflammation-related targets through the DisGeNET database,etc.Use Cytoscape 3.7.2 software to construct the network diagram of drug-active components-targets.Enter potential targets into the STRING platform to build a protein interaction(PPI)network between targets.The DAVID database was used for GO enrichment analysis and KEGG pathway enrichment analysis for potential targets.Results A total of 8 active ingredients including myristic acid,palmitic acid,palmitoleic acid,and 208 corresponding targets were screened.Through Venny 2.1.0,19 components such as PPARG,PPARA,PTPN1,PTGES,RORC and the intersection of disease targets were screened out,and the signal pathways involved include cancer pathways,non-alcoholic fatty liver(NAFLD),etc.Conclusion This study found that sea buckthorn oil regulates inflammatory response through multiple components and multiple targets,which provided guidance for follow-up research.
