摘  要:目的观察肌腱蛋白C(TNC)、肌腱蛋白X(TNX)、合成型平滑肌细胞(SMC)标记蛋白原肌球蛋白4(TPM4)与动脉粥样硬化(AS)斑块形成的关系。方法选择SPF级雄性载脂蛋白E敲除小鼠40只为实验组,高脂饲料喂养;选择雄性C57BL/6小鼠30只为对照组,普通饲料喂养。检测2组血脂,采用免疫组织化学染色检测TNC、TNX及TPM4表达。结果实验组喂养10、16、22、28、34周体质量、TC、LDL,喂养34周TG,喂养22、28、34周HDL水平明显高于对照组(P<0.05,P<0.01)。与16周、22周、28周比较,实验组34周动脉斑块内TNC、TNX及TPM4表达显著增高。AS斑块内TNC、TNX、TPM4表达与斑块面积呈正相关(r=0.585,r=0.915,r=0.863,P<0.05)。TNC与TNX、TNC与TPM4及TNX与TPM4表达呈正相关(r=0.529,r=0.372,r=0.727,P<0.05)。TNC与TPM4、TNX与TPM4相关(P<0.05)。结论在AS斑块形成过程中,随着斑块面积增大,TNC、TNX及合成型SMC数量逐渐增加,TNC与TNX可能存在协同正性调控纤维斑块形成及进展的关系。Objective To study the relationship of TNC,TNX,TPM4 expressions with fibrous plaque formation.Methods Forty male SPF apolipoprotein E-knocked out mice served as an experimental group and 30 male C57 BL/6 mice served as a control group.The blood lipid level was measured and the expressions of TNC,TNX and TPM4 were detected with immunohistochemical staining after the animals in experimental group were fed on a high fat diet and those in control group were fed on a common diet.Results The BMI at weeks 10,16,22,28,34,the serum TC and LDL levels at week 34,and the serum HDL level at weeks 22,28 and 34 were significantly higher in experimental group than in control group(P<0.05,P<0.01).The expression levels of TNC,TNX and TPM4 were significantly higher in experimental group at week 34 than at weeks 16,22 and 28(P<0.05,P<0.01).The expressions of TNC,TNX and TPM4 in atherosclerotic plaques were positively related with plaque size(r=0.585,r=0.915,r=0.863,P<0.05).TNC was positively related with TNX and TPM4 expressions.TNX was positively related with TPM4 expression(r=0.529,r=0.372,r=0.727,P<0.05).TNC and TNX were related with TPM4(P<0.05).Conclusion TNC,TNX and synthetic SMC increase gradually with the increasing plaque size in atherosclerotic plaque formation process.TNC and TNX can synergistically regulate the formation and progression of atherosclerotic fibrous plaques in male SPF mice.
