摘  要:目的探讨结直肠癌组织中结肠癌转移相关基因1(MACC1)和Polo样激酶1(PLK1)表达的临床意义。方法收集2016年4月至2021年3月大庆油田总医院资料完整的结104例结直肠癌组织标本及癌旁正常组织标本,采用免疫组织化学法检测各组织中MACC1蛋白和PLK1蛋白表达,采用χ^(2)检验分析评估MACC1和PLK1的表达水平与结直肠癌临床病理因素之间的关系。结果MACC1蛋白在结直肠癌组织中表达率为73.08%(76/104),明显高于癌旁组织中表达率15.39%(16/104),两者差异有统计学意义(χ^(2)=70.165,P<0.01);PLK1蛋白在结直肠癌组织中表达率为57.70%(60/104),明显高于癌旁组织中表达率24.04%(25/104),两者差异有统计学意义(χ^(2)=24.371,P<0.01)。MACC1表达水平与结直肠癌淋巴结转移、浸润深度、TNM分期明显相关(χ^(2)=4.697、6.307、6.899,P<0.05),PLK1表达水平与结直肠癌淋巴结转移、组织学分化程度、TNM分期明显相关(χ^(2)=5.446、5.5076、6.307,P<0.05)。结论MACC1蛋白和PLK1蛋白表达与结直肠癌侵袭和转移密切相关,检测上述两种指标,并与临床指标结合,将能更准确判断结直肠癌生物学行为及预后、并指导临床治疗。Objective To investigate the clinical significance of metastasis-associated in colon cancer 1(MACC1)and Polo-like kinase 1(PLK1)expression in colorectal carcinoma tissue.Methods A total of 104 cases of colorectal carcinoma and adjacent normal tissue samples with complete data were collected from Daqing Oilfield General Hospital from April 2016 to March 2021,the expression of MACC1 protein and PLK1 protein in each tissue was detected by immunohistochemical method,Chi-square test was used to evaluate the relationship between the expression levels of MACC1 and PLK1 and clinicopathological factors in colorectal carcinoma.Results The expression rate of MACC1 protein in colorectal carcinoma tissues was 73.08%(76/104),which was significantly higher than that in paracancerous tissues 15.39%(16/104),the difference was statistically significant(χ^(2)=70.165,P<0.01),the expression rate of PLK1 protein in colorectal carcinoma tissues was 57.70%(60/104),significantly higher than that in paracancerous tissues 24.04%(25/104),the difference was statistically significant(χ^(2)=24.371,P<0.01).The expressions of MACC1 were significantly correlated with lymph node metastasis,invasion depth,and tumor node metastasis stage(TNM stage)of colorectal carcinoma(χ^(2)=4.697,6.307,6.899,P<0.05),the difference was statistically significant.The expressions of PLK1 were significantly correlated with lymph node metastasis,histology grade and TNM stage of colorectal carcinoma(χ^(2)=5.446,5.508,6.307,P<0.05),the difference was statistically significant.Conclusion The expression of MACC1 protein and PLK1 protein were closely related to the invasion and metastasis of colorectal carcinoma,detection of these two indexes combined with clinical indicators,will be able to more accurately determine the biological behavior and prognosis of colorectal carcinoma and guide the clinical treatment.
