摘 要:目的:探讨有丝分裂相关蛋白TPX2、Aurora-A、FBXW7在食管癌发生发展中的作用及预后机制。方法:选取127例食管癌患者的癌组织与癌旁组织标本为癌组织组及癌旁组织组;另选取60例食管高级别上皮内瘤变组织标本为对照组。运用免疫组化检测三组TPX2、Aurora-A、FBXW7蛋白表达情况;分析TPX2、Aurora-A、FBXW7表达与食管癌患者各项临床病理特征的相关性;通过Kaplan-Meier生存分析法分析患者5年生存率与TPX2、Aurora-A、FBXW7表达的关系。结果:癌组织组TPX2、Aurora-A、FBXW7蛋白阳性率高于癌旁组织组及对照组(P<0.05);Aurora-A蛋白表达水平与肿瘤浸润深度和淋巴结转移密切相关(P<0.05);FBXW7在癌组织组中的表达水平低于癌旁组织组和对照组(P<0.05),其与食管癌患者各项病理特征均无明显相关性(P>0.05);Aurora-A与TPX2表达呈正相关(P<0.05),与FBXW7表达呈负相关(P<0.05);TPX2与FBXW7的表达无明显相关性(P>0.05)。生存分析示,TPX2与食管癌患者5年生存率相关(P<0.05);Aurora-A与FBXW7与患者预后无明显相关性(P>0.05)。结论:食管癌发生发展中,有丝分裂相关蛋白TPX2、Aurora-A、FBXW7均有参与,而在其预后判断中,TPX2具有更高的应用价值。Objective:To investigate the mechanism of mitotic related proteins TPX2,Aurora-A and FBXW7 in the pathogenesis and development of esophageal carcinoma and their prognostic mechanisms.Methods:The cancer tissues and adjacent tissues of 127 patients with esophageal cancer were selected as cancer tissue group and adjacent tissue group,another 60 cases of esophageal high-grade intraepithelial neoplasia were selected as the control group.The expression of TPX2,Aurora-A and FBXW7 protein in the three groups were detected by immunohistochemistry,and the correlation between the expression of TPX2,Aurora-A and FBXW7 and the clinicopathological features of patients with esophageal cancer were analyzed.The relationship between 5-year survival rate and the expression of TPX2,Aurora-A and FBXW7 were analyzed by Kaplan-Meier survival analysis.Results:The positive rates of TPX2,Aurora-A and FBXW7 in cancer tissue group were higher than those in adjacent tissue group and control group(P<0.05).The expression level of Aurora-A protein was closely related to the depth of tumor invasion and lymph node metastasis(P<0.05).The expression of FBXW7 in cancer tissue group was significantly lower than that in adjacent tissue group and control group(P<0.05),it had no significant correlation with the pathological features of esophageal cancer patients(P>0.05).Aurora-A was positively correlated with TPX2 expression(P<0.05),there was a negative correlation between Aurora-A and FBXW7(P<0.05).There was no significant correlation between TPX2 and FBXW7(P>0.05).Survival analysis showed that TPX2 was significantly correlated with 5-year survival rate of patients with esophageal cancer(P<0.05),but Aurora-A and FBXW7 were not significantly correlated with prognosis(P>0.05).Conclusion:Mitosis-related proteins TPX2,Aurora-A and FBXW7 are involved in the occurrence and development of esophageal cancer,and TPX2 has a higher value in the prognosis of esophageal cancer.
