摘　　要：目的探讨肝癌微血管浸润相关长链非编码RNA(lncRNA MVIH)对结直肠癌细胞SW620增殖、侵袭和成瘤能力的影响。方法将体外培养的SW620细胞分为空载体组(转染pcDNA3.1空载体质粒)、MVIH过表达组(转染pcDNA3.1-MVIH过表达载体质粒)、NC-siRNA组(转染干扰序列阴性对照NC-siRNA)和MVIH-siRNA组(转染MVIH-siRNA干扰序列),采用实时荧光定量PCR(qRT-PCR)法检测各组SW620细胞中MVIH mRNA表达水平,细胞计数试剂盒-8(CCK-8)法、克隆形成实验和Transwell小室分别检测SW620细胞增殖活力、克隆形成能力和侵袭能力;将转染48 h后的各组SW620细胞采用皮下注射的方法构建裸鼠皮下移植瘤,每5天测量移植瘤体积评价肿瘤生长情况,25 d后处死裸鼠测量肿瘤重量。结果MVIH过表达组SW620细胞中MVIH mRNA表达水平、细胞增殖活力和克隆形成率及侵袭细胞数明显高于空载体组(P<0.05);MVIH-siRNA组SW620细胞中MVIH mRNA表达水平、细胞增殖活力和克隆形成率明显降低,且侵袭细胞数明显低于NC-siRNA组(P<0.05)。裸鼠皮下移植瘤实验结果显示,MVIH过表达组裸鼠移植瘤体积、肿瘤重量高于空载体组(P<0.05);MVIH-siRNA组裸鼠移植瘤体积、肿瘤重量低于NC-siRNA组(P<0.05)。结论LncRNA MVIH具有促进结直肠癌SW620细胞增殖、侵袭和成瘤的作用。Objective To investigate the effects of IneRNA associated with microvascular invasion in hepatoellular carcinoma(IncRNA MVIH)on proliferation,invasion and tumorigenic ability of colorectal cancer SW620 cells.Methods SW620 cells culured in riro were divided into empty vector group,(transfected with peDNA3.I empty vector plasmid),MVIH overexpression group(transfected with peDNA3.1-MVIH overexpression vector plasmid),NC siRNA group(transfected with interference sequence negative control NC siRNA)and MVIH siRNA group(transfected with MVIH siRNA interference sequence).The mRNA expression level of MVIH in SW620 cells in each group was detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).Cell counting kit-8(CCK-8),clone formation test and transwell chamber were used to detect the proliferation activity,clone formation ability and invasion ability of SW 620 cells;The subcutaneous transplanted tumor of nude mice was constructed by subeutaneous injection of SW620 cells in each group 48 hours after transfection.The transplanted tumor volume was measured every 5 days to evaluate the tumor growth.On the 25th day,the nude mice in each group were killed and the tumor weight was measured.Results The mRNA MVIH expression level,cell proliferation,clone formation rate and number of invasive cells of SW620 cells in the MVIH overexpression group were significantly higher than those in the emply vector group(P<0.05);the MVIH mRNA expression level,cell proliferation,clone formation rate and number of invasive cells of SW620 cells in the MVIH-siRNA group were significantly lower than those in NC-siRNA group(P<0.05).The results of subcutaneous transplantation tumor in nude mice showed that the growth rate of tumor in MVIH overexpression group was faster and the tumor weight was higher than those in the emply vector group(P<0.05);the growth rate of turmor in MVIH overexpression group was slower and the tumor weight was lower than those in NC-siRNA group(P<0.05).Conclusions LncRNA MVIH can promote the prolifer
