摘　　要：目的:探讨沉默Galectin-3对人甲状腺乳头状癌细胞增殖、侵袭能力的影响及其主要机制。方法:分为空白对照组(Control,无特殊处理)、阴性干扰组NC-siRNA(非特异性siRNA)、RNA干扰组(人甲状腺乳头状癌细胞中转染干扰Galectin-3表达的siRNA1及siRNA2),使用MTT法分别检测转染12、24及48 h细胞增殖率;使用Western-blot方法检测转染48 h后各组Galectin-3表达情况及对bcl-2、survivin、MMP-9表达的影响;Transwell小室检测细胞侵袭能力。结果:沉默Galectin-3后人甲状腺乳头状癌细胞的Galectin-3表达与空白对照组及阴性对照组相比明显降低(P<0.05),差异有统计学意义;细胞增殖、侵袭能力明显下降(P<0.05),差异有统计学意义;Bcl-2、Survivin、MMP-9蛋白表达显著低于空白对照组及阴性对照组(P<0.05),差异有统计学意义。结论:沉默Galectin-3基因表达可以降低人甲状腺乳头状癌细胞增殖及侵袭能力,该机制可能与其降低bcl-2、Survivin及MMP-9蛋白表达有一定相关性。Objective:To study the effect of silencing Galectin-3 on the proliferation and migration ability of thyroid cancer cells,and explore the possible mechanism.Methods:There were control group(no speicial treatment)、NC-siRNA group(non-specific siRNA),RNA interference group(transfected into human thyroid carcinoma B-CPAP cells,siRNA1 and siRNA2).MTT was adopted to detect cell proliferation rate after cells treatment in 12,24 and 48h;the Galectin-3、Bcl-2、Survivin and MMP-9 expression were detected by Western blot after 48 h;the cellular migration ability were detected by Transwell chamber test.Results:After silencing Galectin-3 gene in thyroid cancer cells the expression level of Galectin-3 protein in Galectin-3-siRNA1 and Galectin-3-siRNA2 were lower than that in control group and negative group(P<0.05),and the differences were statistically significant.The ability of proliferation and migration decreased significantly than that in the control group and negative group(P<0.05);the expression of Bcl-2,Survivin and MMP-9 protein in B-CPAP cells were significantly lower than those in the control group and negative group(P<0.05).Conclusion:Silencing Galectin-3 gene expression can reduce the proliferation and invasion of B-CPAP,which may be related to the decrease of Bcl-2,survivin and MMP-9 protein expression.
