摘  要:目的:构建肿瘤坏死因子(TNF-α)诱导的足细胞损伤模型,观察不同浓度、相同浓度和不同诱导时间对足细胞Synaptopodin、14-3-3β、RhoA相关蛋白的影响。方法:将0 ng/mL,5 ng/mL,10 ng/mL,20 ng/mL,30 ng/mL,40 ng/mL浓度的TNF-α加入已分化培养6.5 d的足细胞中,诱导培养24 h;给予相同浓度的TNF-α10 ng/ml作用时间分别为6、12、24、36、48 h;给予相同浓度的TNF-α20 ng/ml,作用时间分别为6、12、24、36、48 h。通过Western Blot检测足细胞相关蛋白Synaptopodin、14-3-3β、RhoA的表达,鉴定足细胞的损伤。结果:足细胞在加入10 ng/mL TNF-α时,足细胞开始发生损伤,给予TNF-α浓度为10 ng/mL和20 ng/mL,作用6 h时足细胞均已经发生损伤,且随着作用时间越长,损伤越严重。在损伤足细胞中Synaptopodin、14-3-3β、RhoA蛋白表达下调。结论:随着TNF-α作用浓度及作用时间的增加,足细胞受损严重,相关蛋白表达量明显降低。Objective:To build the tumor necrosis factor(TNF-a)induced cell injury model,and to observe the influence of different concentration,the same concentration and different induced time on Synaptopodin,14-3-3β,and RhoA related proteins of podocytes.Methods:0 ng/mL,5 ng/mL,10 ng/mL,20 ng/mL,30 ng/mL,40 ng/mL concentration of TNF-a was added into the diversified trained 6.5 d cells,with induction training for 24 h;The same concentration of TNF-a 10 ng/ml was given respectively for 6,12,24,36 and 48 h.The same concentration of TNF-a 20 ng/ml was given respectively for 6,12,24,36 and 48 h.Western Blot detection was used to detect podocyte-related Synaptopodin,14-3-3β,and the expression of RhoA to identify podocyte injury.Results:With 10 ng/mL TNF-a,podocyte began to have damage.With TNF-a 10 ng/mL and TNF-a 20 ng/mL for 6 hours,podocyte injury occurred.The longer the time was,the more serious the damage.In the injured podocyte Synaptopodin,14-3-3βand RhoA protein expression decreased.Conclusion:With the increase of TNF-a concentration and action time,podocyte damage will be serious,and the related protein expression quantity decreases obviously.
