摘 要:目的:探讨褪黑素对内蒙古绒山羊毛囊干细胞增殖和迁移的影响及机制。方法:将培养后的内蒙古绒山羊毛囊干细胞分为空白对照组、褪黑素干预A组、褪黑素干预B组、褪黑素干预C组、IWR-1组、褪黑素+IWR-1组,利用CCK8实验检测各组细胞增殖活性,Transwell细胞迁移实验检测各组细胞的迁移数目。经Western Blot法检测各组LEF-1、β-catenin蛋白相对表达量。结果:伴随褪黑素浓度的增加,毛囊干细胞的活力值、迁移细胞数目有显著升高趋势。IWR-1组的细胞活力值、迁移细胞数目均显著低于褪黑素干预组(P<0.05);褪黑素+IWR-1组的细胞活力值、迁移细胞数目均显著高于IWR-1组(P<0.05)。Western-blot检测结果显示,褪黑素干预组的β-catenin蛋白、LEF-1蛋白的相对表达量明显高于空白对照组(P<0.05);IWR-1组的β-catenin蛋白、LEF-1蛋白的相对表达量显著低于褪黑素干预组(P<0.05);褪黑素+IWR-1组的β-catenin蛋白、LEF-1蛋白的相对表达量显著高于IWR-1组(P<0.05)。结论:褪黑素可能通过激活典型的Wnt/β-catenin通路促进毛囊干细胞的增殖与迁移。Objective:To investigate the effect and mechanism of melatonin on the proliferation and migration of hair follicle stem cells of Inner Mongolia cashmere goats.Methods:The cultured hair follicle stem cells of Inner Mongolia cashmere goats were divided into blank control group,melatonin intervention group A,melatonin intervention group B,melatonin intervention group C,IWR-1 group,and melatonin+IWR-1 group.The proliferation activity of each group was detected by CCK8 assay.Transwell cell migration assay was used to detect the number of cells in each group.The relative expression levels of LEF-1 andβ-catenin in each group were detected by Western Blot.Results:With the increase of melatonin concentration,the activity value of hair follicle stem cells and the number of migratory cells increased significantly.The cell viability value and the number of migrated cells in IWR-1 group were significantly lower than those in melatonin intervention group(P<0.05).The cell viability value and number of migrated cells in melatonin+IWR-1 group were significantly higher than those in IWR-1 group(P<0.05).Western-blot results showed that the relative expression levels ofβ-catenin protein and LEF-1 protein in melatonin intervention group were significantly higher than those in blank control group(P<0.05).The relative expression levels ofβ-catenin protein and LEF-1 protein in IWR-1 group were significantly lower than those in melatonin intervention group(P<0.05).The relative expression levels ofβ-catenin protein and LEF-1 protein in melatonin+IWR-1 group were significantly higher than those in IWR-1 group(P<0.05).Conclusion:Melatonin may promote the proliferation and migration of hair follicle stem cells by activating the typical Wnt/β-catenin pathway.
