摘 要:利用反胶束萃取法对紫花芸豆凝集素进行分离纯化,经响应面优化试验得到最佳参数:前萃取离子强度50 mmol/L,萃取时间20 min,前萃取pH 5.8, AOT浓度60 mmol/L,后萃取pH 9.0,后萃取离子强度530 mmol/L。在该条件下,蛋白回收率为82.66%,纯化倍数为9.65倍,试验证明反胶束萃取可用于紫花芸豆凝集素的分离纯化并保留其活性。The lectin of Purple kidney bean was isolated and purified by reverse micelle extraction, and its physical and chemical properties were determined. The optimal parameters obtained by the response surface optimization test were as follows: pre-extraction ion concentration was 50 mmol/L, extraction time was 20 min, pre-extraction pH was 5.8, AOT concentration was 60 mmol/L, post-extraction pH was 9.0, and post-extraction ion concentration was 530 mmol/L. Under this condition, the protein recovery rate was 82.66%, and the purification rate was 9.65 times, which proved that reverse micelle extraction could be used to isolate and purify lectin from purple kidney bean and retain its activity.
