摘 要:目的 :研究淫羊藿苷(ICA)对骨质疏松大鼠牙种植体骨整合的影响,并探讨其可能的机制。方法 :将60只大鼠随机分为假手术组、模型组、ICA组、ICA+Noggin组,每组15只。模型组、ICA组、ICA+Noggin组建立骨质疏松模型,假手术组仅行"假"手术,所有大鼠行单侧胫骨干骺端钛种植体植入术。ICA组以ICA 25 mg/kg灌胃,ICA+Noggin组以ICA 25 mg/kg、Noggin 75 ng/kg灌胃,假手术组、模型组以等体积生理盐水灌胃,1次/d,连续12周。采用亚甲基蓝-碱性品红染色法观察种植术后4、12周种植体愈合情况及骨组织形态;Western blot免疫印迹检测种植体周围骨组织骨形态发生蛋白2(BMP-2)、Runt相关转录因子2(Runx2)、成骨相关转录因子抗体(OSX)蛋白表达。采用SPSS 19.0软件包对数据进行统计学分析。结果:与假手术组比较,模型组种植术后4、12周骨结合率、松质骨区骨量,表面骨质厚度,种植体周围骨组织BMP-2、Runx2、OSX蛋白相对表达量均显著降低(P<0.05)。与模型组比较,ICA组种植术后4、12周骨结合率、松质骨区骨量、表面骨质厚度,种植体周围骨组织BMP-2、Runx2、OSX蛋白相对表达量显著升高(P<0.05)。与ICA组相比,ICA+Noggin组种植术后4、12周骨结合率、松质骨区骨量、表面骨质厚度,种植体周围骨组织BMP-2、Runx2、OSX蛋白相对表达量显著降低(P<0.05)。结论:ICA可促进骨质疏松大鼠牙种植体骨整合,其作用机制可能与激活BMP-2/Runx2信号通路有关。PURPOSE: To study the effect of icariin(ICA) on osseointegration of dental implants in osteoporotic rats, and to explore its possible mechanisms. METHODS: Sixty rats were randomly divided into sham operation group, model group, ICA group, ICA+Noggin group, 15 rats in each group. Osteoporosis models were established in the model group, ICA group, and ICA+Noggin group. The sham operation group only underwent sham operation. All rats underwent unilateral tibial metaphysis titanium implant implantation. The ICA group was given ICA 25 mg/kg by gavage, the ICA+Noggin group was given ICA 25 mg/kg and Noggin 75 ng/kg by gavage, and the sham operation group and model group were gavaged with equal volume of normal saline, once a day for 12 consecutive weeks. Methylene blue-basic fuchsin staining method was used to observe implant healing and bone tissue morphology 4 and 12 weeks after implantation. Western blot was used to detect the protein expression of bone morphogenetic protein-2(BMP-2), Runt-related transcription factor 2(Runx2),and osteogenic-related transcription factor antibody(OSX) in the bone tissue around the implant. SPSS 19.0 software package was used for data analysis. RESULTS: Compared with sham operation group, the osseointegration rate, cancellous bone volume, surface bone thickness, and relative expression of BMP-2, Runx2, and OSX protein in the bone tissue around the implant in the model group were significantly decreased 4 and 12 weeks after implantation(P<0.05). Compared with model group, the osseointegration rate, cancellous bone volume, surface bone thickness, and relative expression of BMP-2, Runx2, and OSX protein in the bone tissue around the implant in ICA group were significantly decreased 4 and12 weeks after implantation(P<0.05).
