摘　　要：目的分析肠道病毒71型(EV71)感染对神经胶质瘤细胞谷胱甘肽S-转移酶P-1(GSTP1)基因表达及细胞迁移、侵袭的影响。方法培养神经胶质瘤U251细胞,行EV71感染,分为EV71感染和空白对照组,分析EV71感染对神经胶质瘤细胞GSTP1基因表达及细胞迁移、侵袭的影响。结果光镜下可见,与空白对照组相比,经EV71感染处理48 h后的神经胶质瘤U251细胞形态变化,存在典型的细胞病变效应现象,细胞钝化、变小,细胞间隙增大。聚合酶链反应(PCR)检测结果显示,与空白对照组比较,EV71感染组细胞中GSTP1基因表达量升高,12、24、48 h细胞迁移率和细胞侵袭数均升高,差异均具有统计学意义(P<0.05)。结论 EV71感染可明显增加GSTP1基因在神经胶质瘤细胞中的表达,促进细胞迁移、侵袭,这可能是EV71感染引起神经系统功能紊乱的机制之一。OBJECTIVE To observe the effect of enterovirus 71(EV71) infection on glutathione S-transferase(GSTP1) gene expression, migration and invasion of glioma cells. METHODS U251 glioma cells were cultured and infected with EV71. They were divided into the EV71 infection group and the blank control group. The effects of EV71 infection on GSTP1 gene expression, migration and invasion of glioma cells were analyzed. RESULTS Under the light microscope, compared with the blank control group, the morphology of glioma U251 cells treated with EV71 for 48 h changed significantly, and there were typical cytopathic effects such as cell passivation, getting smaller, and increase of intercellular space. The result of polymerase chain reaction(PCR) showed that the expression level of GSTP1 gene of the EV71 infection group was significantly higher than that of the blank control group;the cell migration rate and number of invaded cells were significantly higher in the EV71 infection group than in the blank control group(P<0.05). CONCLUSION EV71 infection can significantly increase the expression of GSTP1 gene in glioma cells and promote cell migration and invasion, which may be one of the mechanisms that the EV71 infection leads to dysfunction of nervous system.
