摘　　要：【目的】分析钙蛋白酶1(CAPN_(1))、CAPN_(2)在喉鳞状细胞癌组织中的表达与患者临床病理特征的关系,探讨二者在喉癌发生发展中的作用。【方法】应用酶联免疫吸附法检测65例喉癌患者、60例声带息肉患者血清中CAPN_(1)、CAPN_(2)的含量,应用荧光定量PCR法和蛋白质印迹法检测65例喉癌组织、癌旁组织及60例声带息肉组织中CAPN_(1)、CAPN_(2)、NF-κB、IκB的表达情况,并分析CAPN_(1)和CAPN_(2)的表达与喉鳞状细胞癌临床病理因素的关系。【结果】与声带息肉组相比,喉鳞状细胞癌组血清CAPN_(1)、CAPN_(2)、癌组织CAPN_(1)mRNA、CAPN_(2)mRNA、NF-κB、癌旁组织NF-κB均升高(P<0.05)。CAPN_(1)、CAPN_(2)的表达与淋巴结转移、肿瘤分期、病理分级有关(P<0.05)。【结论】CAPN_(1)、CAPN_(2)的过表达在喉癌的发生、发展和转移中有重要作用,可能是喉鳞状细胞癌诊断和治疗的新靶标。【Objective】To study the relationshipof the expression of calpain(CAPN)1 and CAPN_(2)in laryngeal squamous cell carcinoma and the clinicopathological features of patients,investigate the role of CAPN_(1)and CAPN_(2)in the occurrence and development of laryngeal carcinoma.【Methods】The levels of CAPN_(1)and CAPN_(2)in serum of 65 patients with laryngeal carcinoma and 60 patients with vocal cord polyps were detected by enzyme-linked immunosorbent assay(ELISA),and CAPN_(1),CAPN_(2),NF-κB and IκB in 65 cases of laryngeal carcinoma,paracancerous tissues and 60 cases of vocal cord polyps were detected by PCR and Western blot.The relationship between theexpression of the CAPN_(1)and CAPN_(2)and clinicopathological features of laryngeal squamous cell carcinoma was analyzed.【Results】Compared with vocal cord polyp group,serum CAPN_(1),CAPN_(2),cancer tissue CAPN_(1)mRNA,CAPN_(2)mRNA and NF-κB,adjacent tissue NF-κB in laryngeal squamous cell carcinoma group increased(P<0.05).The expression of CAPN_(1)and CAPN_(2)was related to lymph node metastasis,tumor stage and pathological grade(P<0.05).【Conclusion】The overexpression of CAPN_(1)and CAPN_(2)plays an important role in the occurrence,development and metastasis of laryngeal squamouscell carcinoma,and may be a new target for judging and treatment of laryngeal squamous cell carcinoma.
