摘 要:目的:探究miR-154通过调控N-乙酰氨基半乳糖转移酶7(GALNT7)对人脑星形细胞瘤(U251)细胞的增殖、迁移及侵袭产生影响。方法:将U251细胞设置为对照组、miR-NC组、miR-154 mimics组,采用RT-qPCR检测人脑星形细胞瘤(U87、U251、SHG44)细胞及人脑胶质(HEB)细胞中miR-154的表达情况;MTT法和平板克隆法检测各组U251细胞增殖情况;Transwell法检测各组U251细胞侵袭、迁移情况;双荧光素酶报告基因检测miR-154与GALNT7的靶向关系;western blotting法检测各组细胞GALNT7及增殖、侵袭迁移相关蛋白表达情况。结果:与HEB人脑胶质细胞相比,人脑星形细胞瘤U87、U251、SHG44细胞中miR-154表达水平均显著降低,并且在U251细胞中其表达量最低(P<0.05)。与对照组比较,miR-NC组U251细胞存活率、细胞克隆数、侵袭和迁移数、c-Myc、CyclinD1、MMP9、MMP-2表达水平无明显变化(P>0.05),miR-154 mimics组细胞存活率、细胞克隆数、侵袭和迁移数、c-Myc、CyclinD1、MMP9、MMP2表达水平显著降低(P<0.05)。靶基因预测结果显示GALNT7是miR-154的潜在靶基因,二者存在靶向关系。结论:过表达miR-154可能通过下调GALNT7表达抑制U251细胞增殖、侵袭和迁移。Objective:To explore the effect of miR-154 on the proliferation,migration and invasion of human astrocytoma(U251) cells by regulating N-acetylgalactosaminyltransferase 7(GALNT7).Methods:U251 cells were divided into control group,miR-NC group and miR-154 mimics group.The expression of miR-154 in human astrocytoma cells(U87,U251,SHG44) and human glioma cells(HEB) was assessed by RT-qPCR.The proliferation,invasion and migration of U251 cells were detected by MTT assay,plate cloning assay,Invasion and migration of U251 cells were detected by Transwell assay.The targeting relationship between miR-154 and GALNT7 was verified by double luciferase reporter genes assay.The expressions of GALNT7,proliferation,invasion and migration related proteins were determined by western blotting.Results:Compared with HEB cells,the expression level of miR-154 in U87,U251 or SHG44 cells was significantly decreased,and that was the lowest in U251 cells(P<0.05).The cell survival rate,the number of cloning cells,the number of migrated and invasive cells,and the expression levels of c-Myc,CyclinD1,MMP9 and MMP2 were significantly decreased in miR-154 mimics group(P<0.05).There was no significant different between miR-NC group and control group(P>0.05).GALNT7 was a potential target gene of miR-154.Conclusion:Overexpression of miR-154 may inhibit the proliferation,invasion and migration of U251 cells by down-regulating the expression of GALNT7.
