摘 要:目的:探讨白藜芦醇(RSV)调控金属蛋白酶9(ADAM9)基因对宫颈癌细胞增殖和迁移能力的影响,并探讨其作用的可能机制。方法:常规培养宫颈癌细胞HeLa,HeLa经不同浓度的RSV处理后采用CCK8实验检测细胞增殖能力。将HeLa细胞分为NC组和RSV组,Transwell实验检测各组细胞迁移能力,western blotting检测各组细胞中ADAM9蛋白的表达,流式细胞仪检测各组细胞活性氧(reactive oxygen species,ROS)的产生;构建ADAM9敲减质粒和ADAM9过表达质粒,将宫颈癌细胞HeLa分为RSV组、RSV+NC组、RSV+sh-ADAM9组和RSV+oe-ADAM9,各组质粒转染48 h后,采用CCK8实验、Transwell实验、流式细胞仪实验分别检测各组细胞增殖能力、迁移能力和ROS的产生。结果:RSV呈浓度依赖性的抑制宫颈癌细胞HeLa的增长(P<0.05)。与NC组相比,RSV组细胞转移能力下降、ADAM9蛋白的表达减少、ROS产量减少(P<0.05)。CCK8和Transwell实验显示,ADAM9敲减质粒增加RSV对宫颈癌细胞增殖和迁移能力及ROS的抑制作用,ADAM9过表达质粒减弱RSV对宫颈癌细胞增殖和转移能力及ROS产生的抑制作用(P<0.05)。结论:RSV可能通过调控ADAM9基因抑制宫颈癌细胞增殖和迁移能力。Objective:To study the effect and mechanism of resveratrol(RSV) regulating disintegrin and metalloproteinase domain-containing protein 9(ADAM9) on the proliferation and migration of cervical cancer cells.Methods:Cervical cancer HeLa cells were conventionally cultured and treated with different concentrations of RSV.The cell proliferation was detected by CCK8 assay.The HeLa cells were divided into NC group and RSV group.The migration of cells was measured by Transwell assay.The protein expression of ADAM9 was determined by western blotting.The production of reactive oxygen species(ROS) was detected by flow cytometry.The ADAM9 knockdown plasmid and ADAM9 overexpression plasmid were constructed,The HeLa cells were then divided into RSV group,RSV+NC group,RSV+sh-ADAM9 group and RSV+oe-ADAM9 group.After 48 h of transfection,the proliferation,migration and ROS production of cells were evaluated as well.Results:RSV inhibited the growth of HeLa cells in a concentration-dependent manner(P<0.05).Compared with NC group,the cell migration,ADAM9 protein expression,and ROS production of cells in RSV group were decreased(P<0.05).ADAM9 knockdown led to enhancements in the inhibitory effects of RSV on the proliferation,migration and ROS production of cervical cancer cells,whereas overexpression of ADAM9 had opposite effects(P<0.05).Conclusion:RSV may inhibit the proliferation and migration of cervical cancer cells via reducing the ADAM9 expression.
