摘　　要：目的探讨胃癌组织中STAT活化抑制蛋白1(PIAS1)表达及意义。方法选取2019年9月至2021年3月间我院经胃镜检查的120例胃癌组织标本和40例非萎缩性胃炎组织标本,采用免疫组化染色观察标本组织中PIAS1表达水平。构建重组腺病毒载体AD5-F35-PIAS1、AD5-F35-PIAS1-siRNA和AD5-F35-Null,使用白细胞介素-6(IL-6)(100ng/ml)条件培养基模拟胃癌炎症微环境,分别转染人胃癌细胞株MGC-803,分为IL-6组、空载体组、PIAS1上调组和PIAS1下调组。结果检测结果显示,非萎缩性胃炎组织中未见PIAS1阳性表达,低分化胃组织中PIAS1低表达,中高分化胃组织中PIAS1高表达。PIAS1下调组胃癌细胞增殖率增高;PIAS1下调组迁移、侵袭的胃癌细胞数增多,差异均有统计学意义(均P<0.05)。PIAS1下调组胃癌细胞中钙粘附蛋白E表达水平较IL-6组和空载体组明显下降,波形蛋白和p-P38MAPK表达水平明显提升,差异均有统计学意义(均P<0.05)。结论PIAS1表达水平与胃癌组织恶性程度呈负相关;在炎症微环境下,PIAS1高表达可通过抑制胃癌细胞发生上皮―间质转化来降低细胞的增殖、迁移和侵袭能力。Objective To explore the expression and significance of protein inhibitor of activated STAT1(PIAS1)in gastric cancer tissues.Methods From September 2019 to March 2021,120 gastric cancer tissue specimens and 40 non-atrophic gastritis tissue specimens were collected at Xi’an People’s Hospital.The expression of PIAS1 was observed using immunohistochemical staining.The recombinant adenovirus vectors AD5-F35-PIAS1,AD5-F35-PIAS1-siRNA and AD5-F35-Null were constructed and transfected into human gastric cancer cell line MGC-803 using interleukin-6(IL-6)(100 ng/ml)conditioned medium.The cells were divided into an IL-6 group,an empty vector group,an up-regulated PIAS1 group and a downregulated PIAS1 group.Results Test results showed that there was no positive expression of PIAS1 in nonatrophic gastritis tissues.PIAS1 was low in poorly differentiated gastric tissues and high in moderately and highly differentiated gastric tissues.The proliferation rate of gastric cancer cells was significantly high in the down-regulated PIAS1 group and the number of migrating and invading gastric cancer cells increased in the down-regulated PIAS1 group(all P<0.05).The expression level of epithelial cadherin(E-cadherin)in gastric cancer cells decreased while the expression levels of vimentin and p-P38 MAPK increased in the down-regulated PIAS1 group compared with the IL-6 group and empty vector group(all P<0.05).Conclusion The expression of PIAS1 is negatively associated with the malignant degree of gastric cancer.In inflammatory microenvironment,high expression of PIAS1 can reduce the proliferation,migration and invasion of gastric cancer cells by inhibiting epithelial-mesenchymal transition.
