摘  要:目的:揭示E2F1在肿瘤中重要的免疫调节因子的地位,为肿瘤的免疫治疗和预后预测提供新的靶点。方法:利用ONCOMINE数据库、UCSC Xena数据库综合分析E2F1在肿瘤组织以及癌旁组织中的表达。在GEPIA数据库中,测试E2F1表达和泛癌分期之间的相关性以及泛癌组织和癌旁组织之间E2F1的表达水平差异。Kaplan-Meier Plotter分析E2F1在泛癌中的差异生存曲线。并综合分析各个肿瘤中E2F1与肿瘤突变负荷(TMB)和微卫星不稳定性(MSI)的相关性。随后使用TISIDB数据库重点研究了每种肿瘤中E2F1与肿瘤免疫浸润细胞之间的相关性。结果:E2F1在多种肿瘤中普遍高表达,且E2F1可以预测多种肿瘤患者的生存率。在多种肿瘤中,E2F1与肿瘤突变负荷和微卫星不稳定性明显相关。E2F1和ACT-CD4淋巴细胞的关系最为明显,尤其是在乳腺浸润癌(BRCA)、肾透明细胞癌(KIRC)和肝细胞肝癌(LIHC)这三种肿瘤中。结论:E2F1是至关重要的肿瘤预后预测因子,并在一定程度上调节多种肿瘤相关的免疫反应。Objective:To reveal the status of E2 F1 as an important immunomodulatory factor in human tumors,and to provide a new target for the immunotherapy and prognosis of human tumors.Methods:The expression of E2 F1 in tumor tissues and adjacent tissues was analyzed by using ONCOMINE and UCSC Xena databases.In the GEPIA database,the correlation between the expression of E2 F1 and the stage of pancarcinoma and the difference in the level of E2 F1 expression between pancarcinoma tissue and adjacent tissue were tested.Kaplan-Meier Plotter was used to analyze the differential survival curve of E2 F1 in pan-cancer.The relationship between E2 F1 and tumor mutation burden(TMB) and microsatellite instability(MSI) was analyzed.The association between E2 F1 and tumor-infiltrating cells in each tumor was then focused using the TISIDB database.Results:E2 F1 was overexpressed in many kinds of human tumors and E2 F1 could predict the survival rate of many kinds of tumor patients.E2 F1 was significantly associated with TMB and MSI in a variety of tumors.The association between E2 F1 and ACT-CD4 was most pronounced,especially in BRCA(breast invasive carcinoma),KIRC(kidney renal clear cell carcinoma),and LIHC(liver hepatocellular carcinoma).Conclusion:E2 F1 is an important prognostic factor of tumor,and can modulate various tumor-related immune responses to some extent.
