摘 要:目的分析番茄病果中交链孢病原菌的分离鉴定及产毒情况。方法采集福建省5个县市番茄病果,通过切片和组织分离法从番茄病害部位分离纯化病原菌,结合形态学特征和分子生物学对病原菌进行鉴定。利用超高效液相色谱-串联质谱法对分离得到的交链孢菌产毒能力进行检测分析。结果在发病番茄病害部位分离得到的3株互隔交链孢菌株均具产毒能力,但毒素种类和产毒能力各有不同。其中,NH06菌株产毒量最高,可产生细交链格孢酮酸(tenuazonic acid,TeA)、交链孢酚(alternariol,AOH)、交链孢酚单甲醚(alternariol monomethyl ether,AME)、交链孢烯(altenuene,ALT)、腾毒素(tentoxin,TEN)5种毒素,而NP002和NH07仅检出产生4种毒素。结论番茄病果中存在多种交链孢菌,其产毒能力存在差异。检测发现番茄病果含链格孢霉毒素时,应及时处理以防成为交链孢菌的污染源,同时应加强产毒病果产地的环境消毒和对同批果品的保护性灭菌。Objective To analyze the isolation,identification and toxin production of Alternaria sp.in tomato diseased fruit.Methods Tomato diseased fruits from 5 counties and cities in Fujian province were collected,the pathogens were isolated and purified from tomato diseased parts by slicing and tissue isolation,the pathogens were identified by morphological characteristics and molecular biology,the toxin producing abilitiees of the isolated Alternaria sp.were detected and analyzed by ultra performance liquid chromatography-tandem mass spectrometry.Results Three strains of Alternaria sp.strains isolated from the diseased site of tomato had the ability to produce toxin,but the types and abilities of toxin production were different.Among them,NH06 strain had the highest toxin production,which could produce 5 kinds of toxins including tenuazonic acid(TeA),alternariol(AOH),alternariol monomethyl ether(AME),altenuene(ALT),and tentoxin(TEN)contains,while NP002 and NH07 strains could only produce 4 kinds of toxins.Conclusion There are many Alternaria sp.in tomato diseased fruit,and their toxin production abilities are different.If Alternaria toxins are found in tomato diseased fruit,it should be treated in time to prevent it from becoming a pollution source of Alternaria sp..At the same time,the environmental disinfection of the producing area of poisonous and diseased fruits and the protective sterilization of the same batch of fruits should be strengthened.
