摘  要:目的探讨活性维生素D_(3)对肾小球系膜细胞中Bcl-2及Bax蛋白表达的影响及调控作用。方法将对数生长期的人肾小球系膜细胞随机分为4组,空白对照组加入MEM正常培养基培养,高糖组加入含30 mmol/L葡萄糖的MEM培养基培养,VD_(3)干预组加入含10^(-8) mol/L 1,25-二羟基维生素D_(3)[1,25-(OH)_(2)D_(3)]的MEM培养基培养,联合干预组加入含10^(-8)mol/L 1,25-(OH)_(2)D_(3)和30 mmol/L葡萄糖的MEM培养基培养。各组均干预48 h后收集细胞,显微镜下观察各组细胞形态,应用流式细胞术检测各组细胞凋亡情况,应用Western blot法检测各组细胞中Bcl-2及Bax蛋白表达情况。结果显微镜下观察,高糖组、VD_(3)干预组、联合干预组细胞数目依次减少,细胞体积缩小、胞核皱缩,可见凋亡小体。高糖组、VD_(3)干预组和联合干预组的细胞凋亡率及Bax蛋白表达量均明显高于空白对照组(P均<0.05),VD_(3)干预组和联合干预组均明显高于高糖组(P均<0.05),联合干预组均明显高于VD_(3)干预组(P均<0.05);高糖组、VD_(3)干预组和联合干预组Bcl-2蛋白表达量及Bcl-2/Bax均明显低于空白对照组(P均<0.05),VD_(3)干预组和联合干预组均明显低于高糖组(P均<0.05),联合干预组明显低于VD_(3)干预组(P均<0.05)。结论高糖干预可促进人肾小球系膜细胞凋亡,活性维生素D_(3)干预高糖诱导下的人肾小球系膜细胞可进一步诱导细胞凋亡,二者诱导细胞凋亡的机制可能与调节凋亡蛋白Bcl-2、Bax的表达有关。Objective It is to explore the effect and regulation of active vitamin D_(3)on the expression of Bcl-2 and Bax protein in glomerular mesangial cells.Methods Human glomerular mesangial cells in the logarithmic growth phase were randomly divided into 4 groups.The blank control group was cultured with MEM normal medium,the high glucose group was cultured with MEM medium containing 30 mmol/L glucose,and the VD_(3)intervention group was cultured with MEM medium containing 10^(-8)mol/L 1,25-dihydroxyvitamin D_(3)[1,25-(OH)_(2)D_(3)],the combined intervention group was cultured with MEM medium containing 10^(-8) mol/L 1,25-(OH)_(2)D_(3)and 30 mmol/L glucose.The cells were collected after 48 hours of intervention in each group,the cell morphology of each group was observed under a microscope,the apoptosis of each group was detected by flow cytometry,and the expression of Bcl-2 and Bax protein in the cells of each group was detected by Western blot.Results Observed under the microscope,the number of cells in the high glucose group,VD_(3)intervention group and combined intervention group decreased successively,the cell volume shrank,the nucleus shrank,and apoptotic bodies were found.The apoptosis rate and Bax protein expression of the high glucose group,the VD_(3)intervention group and the combined intervention group were significantly higher than those of the blank control group(all P<0.05),and the VD_(3)intervention group and the combined intervention group were significantly higher than the high glucose group(all P<0.05),the combined intervention group was significantly higher than the VD_(3)intervention group(P<0.05);the Bcl-2 protein expression and Bcl-2/Bax of the high glucose group,VD_(3)intervention group and combined intervention group were significantly lower than those of the blank control group(P<0.05),and the VD_(3)intervention group and combined intervention group were significantly lower than the high glucose group(all P<0.05),the combined intervention group was significantly lower than the VD_(3)interventio
