葫芦素B抑制CIP2A/AKT表达抗非小细胞肺癌的作用

摘　　要：目的通过观察葫芦素B对非小细胞肺癌A549细胞增殖、细胞周期、细胞迁移及凋亡的影响,及其与CIP2A/AKT信号表达的相关性,明确葫芦素B对非小细胞肺癌的作用及可能机制。方法不同浓度的葫芦素B处理A549细胞不同时间,采用MTS法检测细胞增殖、划痕实验检测细胞迁移、流式细胞仪检测细胞周期及凋亡、Western Blot检测CIP2A、AKT、PAKT表达。结果葫芦素B以时间及浓度依赖性抑制非小细胞肺癌A549细胞的增殖、迁移,对A549细胞存在G_(2)/M期阻滞并促进其凋亡,抑制CIP2A及PAKT表达。结论葫芦素B能够抑制A549细胞增殖、迁移、阻滞细胞周期于G_(2)/M期并促进其凋亡,其作用途径可能为CIP2A/AKT,这为葫芦素B作为CIP2A抑制剂在非小细胞肺癌的治疗中提供了有力证据。Objective To observe the effect of cucurbitacin B on proliferation,cell cycle,migration and apoptosis in non-small cell lung cancer A549 cells,as well as the expression of CIP2A/Akt signal,through which clarifies the role and mechanism of cucurbitacin B in NSCLC.Methods A549 were treated with Cucurbitacin B at different concentrations for different time to perform the following experiments:MTS assay to detect cell viability and proliferation;scratch test to detect the migration ability of cells;flow cytometry to detect cell cycle;flow cytometry to detect apoptosis;The expression of CIP2A,AKT and P-AKT protein was detected by Western Blot.Results Cucurbitacin B inhibited the proliferation and migration of non-small cell lung cancer A549 cells in a time-and concentration-dependent manner,and inhibited the G2/M phase of A549 cells and promoted their apoptosis.Conclusion Cucurbitacin B can inhibit the proliferation,migration,retard cell cycle at G_(2)/M phase and induce apoptosis of A549 cells,its mechanism is CIP2A/AKT pathway.This provides strong evidence for cucurbitacin B as a CIP2A inhibitor in the treatment of non-small cell lung cancer.
