摘  要:目的观察黄芩苷对急性高眼压小鼠视网膜损伤的治疗作用。方法将100只C57BL/6小鼠随机分为正常对照组、高眼压组、黄芩苷25 mg/kg+高眼压组、黄芩苷50 mg/kg+高眼压组、黄芩苷75 mg/kg+高眼压组(每组20只)。其中除正常对照组外,其余各组均通过前房灌注平衡盐溶液建立急性高眼压模型;且在前房灌注前30 min黄芩苷各组均经尾静脉分别注射25、50、75 mg/kg黄芩苷,正常对照组及高眼压组经尾静脉注射生理盐水。造模1、3、5、7 d,用HE染色法观察视网膜组织病理变化并测定视网膜全层厚度,于造模7 d分别用实时荧光定量PCR、Westen blotting法检测视网膜组织中IL-1β、TNF-α、一氧化氮合酶(iNOS)mRNA及蛋白表达。结果与正常对照组比较,高眼压组各时间点视网膜全层厚度均高(P均<0.05),具有时间依赖性;与高眼压组比较,黄芩苷25 mg/kg+高眼压组、黄芩苷50 mg/kg+高眼压组、黄芩苷75 mg/kg+高眼压组各时间点视网膜全层厚度均低(P均<0.05),且具有时间、浓度依赖性。与正常对照组比较,高眼压组视网膜组织中IL-1β、TNF-α及iNOS mRNA及其蛋白表达均高(P均<0.05);与高眼压组比较,黄芩苷25 mg/kg+高眼压组、黄芩苷50 mg/kg+高眼压组、黄芩苷75 mg/kg+高眼压组视网膜组织中IL-1β、TNF-α及iNOS mRNA及其蛋白表达均低(P均<0.05),呈浓度依赖性。结论黄芩苷能有效减轻急性高眼压小鼠视网膜的损伤及炎症反应。Objective To observe the effect of baicalin on retinal injury in mice with acute ocular hypertension.Methods Totally 100 C57BL/6 mice were randomly divided into the normal control group,high intraocular pressure group,baicalin 25 mg/kg+high intraocular pressure group,baicalin 50 mg/kg+high intraocular pressure group,and ba‑icalin 75 mg/kg+high intraocular pressure group,with 20 rats in each group.Except the normal control group,the acute high intraocular pressure models were established by perfusion of balanced salt solution into the anterior cham‑ber;mice in the baicalin groups were injected with 25,50,and 75 mg/kg baicalin through caudal vein 30 min before ante‑rior chamber perfusion,and mice in the normal control group and high intraocular pressure group were injected with normal saline through caudal vein.Hematoxylin eosin(HE)staining was used to observe the histopathological changes of retina and we measured the full thickness of retina at 1,3,5 and 7 days;IL-1β,TNF-α,and iNOS mRNA and protein levels in mouse retina was measured by real-time fluorescence quantitative PCR and Western blotting on the 7th day.Results Compared with the normal control group,the full thickness of retina in the high intraocular pressure group increased in a time-dependent manner(P<0.05).Compared with the high intraocular pressure group,the full thickness of retina in the baicalin 25 mg/kg+high intraocular pressure group,baicalin 50 mg/kg+high intraocular pressure group,and baicalin 75 mg/kg+high intraocular pressure group decreased at each time point(all P<0.05),which was in a time-and concentra‑tion-dependent manner.Compared with the normal control group,IL-1β,TNF-α,and iNOS mRNA and protein in retinal tissues of rats increased in the high intraocular pressure group(all P<0.05).Compared with the high intraocular pressure group,IL-1β,TNF-α,and iNOS mRNA and protein decreased in the retinal tissues of rats in the baicalin 25 mg/kg+high intraocular pressure group,baicalin 50 mg/kg+high intraocular pressure group,and bai
