摘  要:以大果木姜子为试验原料,采用响应面法优化大果木姜子多糖提取工艺,并研究大果木姜子多糖的抗氧化活性。建立以葡萄糖为对照品,紫外分光光度法测定多糖含量的定量分析方法。在单因素试验基础上,以超声时间、提取功率和液料比为自变量,多糖得率为因变量,运用Box-Behnken设计优化大果木姜子多糖的提取工艺。通过大果木姜子多糖对DPPH自由基、超氧阴离子自由基、羟基自由基清除作用,研究其抗氧化活性。结果表明,大果木姜子多糖最佳提取工艺为超声时间35 min、提取功率80 W、液料比40∶1(mL/g),多糖得率为5.92%。大果木姜子多糖对DPPH自由基、超氧阴离子自由基和羟基自由基均有较强的清除能力,在一定浓度范围内,多糖浓度越高,抗氧化活性越强。Taking Cinnamomum migao H.W.Li as the experimental material,the response surface method was used to optimize the extraction process of polysaccharides,and the antioxidant activity of polysaccharides was studied.Establish a quantitative analysis method for the determination of polysaccharide content by ultraviolet spectrophotometry with glucose as the reference substance.On the basis of single factor experiment,taking ultrasonic time,extraction power and liquid-to-material ratio as independent variables,and polysaccharide yield rate as dependent variable.Box-Behnken design was used to optimize the extraction process of polysaccharides from sesame seeds.The anti-oxidant activity of polysaccharides was studied by scavenging DPPH free radicals,superoxide anion free radicals and hydroxyl free radicals.The results showed that the optimal extraction process conditions for polysaccharides were ultrasonic time 35 min,extraction power 80 W,liquid to material ratio 40∶1(mL/g),under these conditions,the polysaccharide yield was 5.92%.The polysaccharides had strong scavenging ability against DPPH free radicals,superoxide anion free radicals and hydroxyl free radicals.Within a certain concentration range,the higher the polysaccharide concentration,the stronger the antioxidant activity.
