摘 要:目的研究中国东北地区人乳头瘤病毒31(HPV31)E6和HPV31 E7基因多态性。方法首先将从已确诊HPV31阳性患者宫颈细胞中提取的DNA,应用巢式PCR对E6和E7基因片段进行扩增并测序,再与参考序列比对找到突变位点。应用MegaX软件进行序列比对以及系统发育树的构建,应用PAML软件分析点突变中的正向选择位点,应用PSIPred server预测HPV31 E6和E7蛋白质二级结构,应用ABCPred server寻找理想的B细胞免疫表位,应用ProPred-Ⅰserver和ProPred server分别预测人类白细胞抗原(HLA)Ⅰ类和HLAⅡ类结合表位。结果成功扩增和测序HPV31 E6和E7序列各151个。与参考序列(J04353.1)比对后发现,E6基因中检测到单核苷酸突变25个,E7基因中检测到单核苷酸突变8个。系统发育分析显示,中国东北地区HPV31 E6和E7基因均以A谱系最为流行。研究观测到的大部分非同义突变存在于预测表位中。结论HPV31 E6和E7基因组在东北地区呈基因多态性分布,并以A谱系为主。Objective To analyze the human papilloma virus(HPV)31 E6 and E7 gene polymorphisms in northeast China.Methods A nested PCR was used to amplify E6 and E7 genetic fragments from DNA extracted from cervical cells of HPV31-positive patients,and mutation sites were detected through sequencing and comparison with reference sequences.Results A total of 151 HPV31 E6 and E7 sequences were successfully amplified and sequenced.After comparison with the reference sequence(J04353.1),25 and 8 single-nucleotide mutations were detected in the E6 and E7 genes,respectively.Phylogenetic analysis determined that lineage A was most commonly observed in both HPV31 E6 and E7 genes.No positive selection sites were detected in all obtained sequences.Most observed non-synonymous mutations were present in the predicted epitopes.Conclusion The HPV31 E6 and E7 genomes are genetically polymorphic among people of northeast China,with lineage A as the most common.
