摘 要:目的观察干扰长链非编码RNA(lncRNA)SNHG22是否通过抑制促红细胞生成素产生肝细胞A型受体3(erythropoietin-producing hepatocellular receptor A3,EPHA3)影响结直肠癌LoVo细胞周期与凋亡。方法利用SNHG22小干扰RNA(si-SNHG22)转染结直肠癌LoVo细胞,构建干扰SNHG22的细胞(si-SNHG22组),转染小干扰RNA阴性对照(si-NC)至LoVo细胞(si-NC组)。实时定量聚合酶链反应(qRT-PCR)检测SNHG22和EPHA3信使RNA(mRNA)的表达水平。流式细胞术评估干扰SNHG22对LoVo细胞周期和凋亡的影响。Western blot检测干扰SNHG22对EPHA3、Cyclin H、Caspase6、CDK3蛋白表达的影响。结果与si-NC组相比,干扰SNHG22显著降低LoVo细胞中EPHA3 mRNA的表达[(0.31±0.06)比(1.31±0.12)](P<0.01),提高G0~G1期的LoVo细胞比例(P<0.01),提高LoVo细胞的凋亡率[(40.61±5.96)%比(10.84±3.22)%](P<0.01)。与si-NC组相比,干扰SNHG22可显著降低LoVo细胞中EPHA3、Cyclin H、CDK3蛋白的表达,显著增加Caspase6蛋白的表达。结论干扰SNHG22可以降低EPHA3基因的表达,阻滞结直肠癌LoVo细胞周期,诱导LoVo细胞凋亡。Objective To observe whether interfering long-chain non-coding RNA(lncRNA)SNHG22 affects the cell cycle and apoptosis of colorectal cancer LoVo cells by inhibiting erythropoietin-producing hepatocellular receptor A3(EPHA3).Methods SNHG22 small interfering RNA(si-SNHG22)was transfected into colorectal cancer LoVo cells to construct cells with low SNHG22 expression(si-SNHG22 group),and small interfering RNA negative control(si-NC)was transfected into LoVo cells(si-NC group).Real-time quantitative polymerase chain reaction(qRT-PCR)was used to detect the expression levels of SNHG22 and EPHA3 messenger RNA(mRNA).Flow cytometry was used to evaluate the effect of interfering SNHG22 on the cell cycle and apoptosis of LoVo cells.Western blot was used to detect the effect of interfering SNHG22 on the protein expressions of EPHA3,Cyclin H,Caspase6,and CDK3.Results Compared with the si-NC group,interference with SNHG22 significantly reduced the expression of EPHA3 mRNA in LoVo cells[(0.31±0.06)vs.(1.31±0.12)](P<0.01),increased the proportion of LoVo cells in the G0-G1 phase(P<0.01),and increased the apoptosis rate of LoVo cells[(40.61±5.96)%vs.(10.84±3.22)%](P<0.01).Compared with the si-NC group,interference with SNHG22 could significantly reduce the protein expressions of EPHA3,Cyclin H,and CDK3 in LoVo cells,and significantly increase the protein expression of Caspase6.Conclusions Interference with SNHG22 can reduce the expression of EPHA3 gene,block the cell cycle of colorectal cancer LoVo cells,and induce the apoptosis of LoVo cells.
