摘 要:目的探究红景天对缺血-低氧(HIE)大鼠血流动力学及神经元凋亡的作用机制。方法共30只,分为假手术组、HIE (建立HIE模型)和红景天组(HIE模型给予红景天灌胃),每组10只。比较三组大鼠血流动力学指标(超声波检测仪检测)、学习能力(Morris水迷宫试验)、神经元凋亡(流式细胞仪检测)、脑组织病理观察(苏木精-伊红染色)及HIF-1 α蛋白(蛋白印迹)差异性。结果与假手术组相比,缺血-低氧组大鼠收缩压(119.18 ± 5.63) mmHg、舒张压(78.41 ± 6.35) mmHg、平均动脉压(93.85±8.63) mmHg及心率(302.49 ± 48.53)次·min^(-1)均明显降低(P <0.05),红景天治疗组大鼠血流动力学指标相比于缺血-低氧组大鼠均有所升高(P<0.05);与假手术组相比,缺血-低氧组各个时间点逃避潜伏期明显升高(P<0.05),治疗1d时,红景天治疗组与缺血-低氧组相比,大鼠逃避潜伏期比较无意义(P>0.05),剩余时间点红景天治疗组大鼠逃避潜伏期均低于缺血-低氧组(均P<0.05);与假手术大鼠海马神经元相比,缺血-低氧组及红景天治疗组神经元凋亡凋亡率明显升高(P<0.01),红景天治疗组相比于缺血-低氧组海马神经元凋亡率明显降低(P<0.01);与假手术组比较,缺血-低氧组神经元细胞中HIF-1 α蛋白表达量明显升高(P<0.05),与缺氧-缺氧组比较,红景天治疗组缺氧诱导因子-1明显降低(P<0.05),红景天治疗组与缺氧诱导因子-1抑制剂组相比,神经元细胞中低氧诱导因子-1蛋白无显著差(P> 0.05)。结论红景天能够改善HIE大鼠血流动力学指标、提高其学习能力,其作用机制可能通过调控HIF-1 α蛋白从而抑制神经元凋亡来实现。Objective To explore the mechanism of Rhodiola on hemodynamics and neuronal apoptosis in ischemia-hypoxia(HIE) rats.Methods A total of 30 animals were divided into sham operation group,HIE(to establish HIE model),and Rhodiola group(HIE model was administered with Rhodiola gavage),with 10 animals in each group.Compare the three groups of rat hemodynamic indicators(detected by ultrasonic detector),learning ability(Morris water maze test),neuronal apoptosis(detected by flow cytometry),brain tissue pathology observation(HE staining) and HIF-1 α Protein(Western blot) differences.Results Compared with the sham operation group,rats in the ischemia-hypoxia group had SBP(119.18 ± 5.63) mmHg,DBP(78.41 ± 6.35) mmHg,MAP(93.85 ± 8.63) mmHg and HR(302.49 ± 48.53)times·min^(-1)were significantly reduced(P <0.05),the hemodynamic indexes of rats in the rhodiola treatment group were increased compared with rats in the ischemia-hypoxia group(P <0.05);compared with the sham operation group Compared with the ischemia-hypoxic group,the escape latency was significantly higher at each time point(P <0.05).At the first day of treatment,compared with the ischemia-hypoxic group,the escape latency of the rats in the rhodiola treatment group was more meaningless(P> 0.05),the escape latency of rats in the rhodiola treatment group was lower than that in the ischemia-hypoxic group(;P <0.05);compared with hippocampal neurons of sham-operated rats,the ischemic-hypoxic group The apoptosis rate of neurons in the Rhodiola treatment group was significantly increased(P <0.01).Compared with the ischemia-hypoxia group,the apoptosis rate of hippocampal neurons in the Rhodiola treatment group was significantly lower(P <0.01);and Compared with the sham operation group,HIF-1α protein expression in neuronal cells in the hypoxia-ischemia group was significantly increased(P <0.05).Compared with the hypoxiahypoxia group,HIF-1α in the rhodiola treatment group was significantly reduced(P <0.05).Compared with the HIF-1α inhibitor group,the rhodiola treatme
