摘 要:目的观察蒙药德都红花-7味散对CCl_(4)所致肝纤维化大鼠Bcl-2、Bcl-x蛋白表达的影响。方法将40只SD雄性大鼠按体重分层随机分为对照组、模型组、水飞蓟素组、德都红花-7味散组,每组10只。采用腹腔注射CCl_(4)方法建立肝纤维化模型,空白对照组和模型组灌胃蒸馏水,德都红花-7味散低剂量组按0.3 g/kg,水飞蓟宾组21 mg/kg灌胃给药,每天给药一次,给药7 w后动物禁食12 h,取肝组织4%多聚甲醛固定。HE染色观察肝组织病理改变,免疫组织化学方法检测大鼠肝组织中Bcl-2、Bcl-x蛋白表达影响。结果HE染色模型组中央静脉、汇管区周围及肝实质内均可见大量的肝细胞脂肪变性,肝细胞内可见体积较小、数量不等的圆形空泡;中等量的汇管区周围可见少量的炎性细胞浸润;小叶结构紊乱,多见纤维间隔形成;未见明显的细胞坏死。水飞蓟宾组、德都红花-7味散组中央静脉、汇管区周围及肝实质内肝细胞颗粒变性明显轻于模型组;小叶结构紊乱,多见纤维间隔形成与模型组比较较轻。免疫组织化学染色与空白组比较模型组Bcl-2、Bcl-x蛋白表达升高,差异有统计学意义(P<0.05);与模型组比较,水飞蓟宾组、德都红花-7味散组Bcl-x蛋白表达下降,德都红花-7味散组Bcl-2蛋白表达下降,差异有统计学意义(P<0.05)。结论蒙药德都红花-7味散能够明显减轻慢性肝损伤,其机制与调节Bcl-2、Bcl-x蛋白表达相关。Objective To observe of the effect of Mongolian medicine DeDuHongHuaQiWeiSan on Bcl-2 and Bcl-x protein expression in rats with CCl_(4)-induced liver fibrosis.Methods Forty MALE SD rats were randomly divided into blank control group,model group,silymarin group and DeDuHongHuaQiWeiSan group according to body weight,with 10 rats in each group.The liver fibrosis model was established by intraperitoneal injection of CCl_(4).The blank control group and the model group were given distilled water intragastric administration,the low-dose DeDuHongHuaQiWeiSan group was given 0.3 g/kg intragastric administration,and the silymarin group was given 21 mg/kg intragastric administration,once a day.7 weeks after administration,the animals were fasting for 12 h,and 4% paraformaldehyde was taken from the liver tissue for fixation.HE staining was used to observe the pathological changes of liver tissue,and the influence of Bcl-2 and Bcl-X protein expression in rat liver tissue was detected by immunohistochemistry.Results A large number of hepatocyte steatosis were observed in the central vein,around the portal area and in the liver parenchyma in the HE staining model group,and small round vacuoles with different numbers were observed in the hepatocytes;A small amount of inflammatory cell infiltration were found around the medium amount of portal area.The lobular structure was disordered,and the formation of fibrous septa was common.The granular degeneration of hepatocytes in central vein,around portal area and in liver parenchyma in silymarin group and DeDuHongHuaQiWeiSan group was significantly lighter than that in model group;the lobular structure was disordered,and the formation of fiber septum was more common than that of model group.Immunohistochemical staining showed the elevated Bcl-2 and Bcl-x protein expression in the model group compared with the blank group,and the difference was statistically significant(P<0.05);Compared with the model group,the protein expression of Bcl-x in silymarin group and DeDuHongHuaQiWeiSan group
