七氟醚调控miR-4458影响胃癌SNU-1细胞的增殖、迁移和凋亡

摘　　要：目的探讨七氟醚是否调控miR-4458影响胃癌SNU-1细胞的增殖、迁移和凋亡。方法采用四甲基偶氮唑蓝(MTT)实验、Transwell实验、流式细胞术、实时定量PCR(RT-qPCR)分析不同浓度七氟醚对SNU-1细胞增殖、迁移、凋亡及miR-4458表达的影响。观察过表达miR-4458或抑制miR-4458表达对SNU-1细胞增殖、迁移和凋亡的影响。结果不同浓度七氟醚处理后SNU-1细胞活力、迁移细胞数显著降低,差异有统计学意义(P<0.05),细胞凋亡率、miR-4458表达显著升高,差异有统计学意义(P<0.05),并呈剂量依赖效应。过表达miR-4458后SNU-1细胞活力、迁移细胞数显著降低(P<0.05),细胞凋亡率显著升高,差异有统计学意义(P<0.05)。抑制miR-4458表达后SNU-1细胞活力、迁移细胞数显著升高,细胞凋亡率显著降低,差异有统计学意义(P<0.05)。抑制miR-4458表达可逆转七氟醚对SNU-1细胞增殖、迁移和凋亡的影响(P<0.05)。结论七氟醚通过上调miR-4458可抑制胃癌SNU-1细胞增殖和迁移,并诱导细胞凋亡。Objective To investigate the effects of sevoflurane on the proliferation,migration and apoptosis of gastric cancer SNU-1 cells by regulating miR-4458.Methods The methyl thiazolyl tetrazolium(MTT)experiment,Transwell experiment,flow cytometry,and real-time quantitative PCR(RT-qPCR)were used to analyze the effects of different concentrations of sevoflurane on the proliferation,migration,apoptosis and miR-4458 expression of SNU-1 cells.Moreover the effects of miR-4458 overexpression or miR-4458 inhibition on the proliferation,migration and apoptosis of SNU-1 cells were detected by the above methods.Results After SNU-1 cells were treated by sevoflurane,the cell viability and migrating cell numbers were significantly decreased(P<0.05),whereas the apoptosis rate and miR-4458 expression were significantly increased(P<0.05),with a dose-dependent manner.After overexpression of miR-4458,the cell viability and the migrating cell numbers of SNU-1 were significantly decreased(P<0.05),however,the apoptosis rate was significantly increased(P<0.05).After the expression of miR-4458 was inhibited,the cell viability and the migrating cell numbers of SNU-1 were significantly increased(P<0.05),whereas the apoptosis rate was significantly decreased(P<0.05).To inhibit the expression of miR-4458 could reverse the effects of sevoflurane on the proliferation,migration and apoptosis of SNU-1 cells(P<0.05).Conclusion Sevoflurane can inhibit the proliferation and migration,and induce cell apoptosis in gastric cancer SNU-1 cells by up-regulating the expression of miR-4458.
