摘  要:目的:探究过敏性哮喘发病过程中关键的基因、信号通路和免疫细胞的浸润情况.方法:从GEO数据库下载数据集GSE63142中表达谱数据.首先,对数据集GSE63142中的原始数据进行背景校检、归一化处理,并使用R语言中的limma包筛选得到差异表达基因.用R语言clusterProfiler包对差异表达基因进行GO分析和KEGG分析,并对原始基因表达矩阵进行GSEA分析.然后通过Cytoscape软件构建差异表达基因的PPI网络,利用插件cytoHubba筛选得到Hub基因,并用GOSemSim包和clusterProfiler包分析了Hub基因的分子相似性.最后利用CIBERSORT探究过敏性哮喘中免疫细胞浸润的情况,并用TIMER数据库验证Hub基因与6种免疫细胞的相关性.结果:通过对基因表达矩阵进行差异表达分析,共得到1 181差异表达基因.GO分析结果表明,过敏性哮喘发病主要与谷胱甘肽衍生物生物合成、谷胱甘肽衍生物代谢过程、核苷酸-糖代谢过程等密切相关.KEGG分析结果显示,炎症介质对TRP通道的调节、IL-17信号通路和肿瘤坏死因子信号通路等在过敏性哮喘的发病过程中可能发挥着重要作用.GSEA分析结果说明,IL-17信号通路、Th1和Th2细胞分化和细胞因子-细胞因子受体相互作用等可能参与了过敏性哮喘的发病过程.通过构建PPI网络共筛选得到10个Hub基因.免疫细胞浸润分析结果表明,过敏性哮喘病人的支气管中出现明显单核细胞、M0巨噬细胞、树突状细胞和中心粒细胞浸润.结论:通过生物信息学分析方法,筛选得到过敏性哮喘发病过程中的关键基因、信号通路以及免疫细胞的浸润情况,从而为过敏性哮喘发病机制的研究提供参考.Objective To explore the key genes, signal pathways and infiltration of immune cells in the pathogenesis of allergic asthma.Methods The expression spectrum data of GSE63142 were downloaded from GEO database.First of all, the original data in GSE63142 were background checked and normalized, and the differentially expressed genes were screened by limma package in R language.The R language cluster profiler package was used to do GO and KEGG analyses on the differentially expressed genes.The gene expression matrix was analyzed using GSEA.Then, the PPI network was constructed by using the software of Cytoscape, and the Hub gene was screened with using the plug-in cytohubba.Both GOSemSim package and clusterProfiler package were used to analyze the molecular similarity of the Hub gene.Finally, CIBERSORT was used to reveal the infiltration of immune cells in allergic asthma, and the TIMER database was used to verify the correlation between the Hub gene and six immune cells.Results 1 181 differentially expressed genes were obtained by differential expression analysis of gene expression matrix.GO analysis showed that the pathogenesis of allergic asthma was closely related to glutathione derivative biosynthesis, glutathione derivative metabolism and nucleotide sugar metabolism.KEGG analysis indicated that the regulation of inflammatory mediators on TRP channel, IL-17 signaling pathway and tumor necrosis factor signaling pathway might be involved in the pathogenesis of allergic asthma.GSEA analysis showed that IL-17 signaling pathway, Th1 and Th2 cell differentiation and cytokine-cytokine receptor interaction may be involved in the pathogenesis of allergic asthma.We screened 10 Hub genes by constructing PPI network.The results showed that monocytes, M0 macrophages, dendritic cells and central granulocytes were infiltrated in the bronchi of patients with allergic asthma.Conclusion Through bioinformatics analysis methods, the key genes, signal pathways and immune cell infiltration in the pathogenesis of allergic asthma are scr
