摘  要:鉴于样品采集与运输过程是影响检测结果能否客观、真实反映生活饮用水中微生物水平的关键步骤,以菌落总数为研究对象,分析样品采集后的保存和运输条件对生活饮用水中微生物检测的影响。结果表明,保存温度、时长以及温度和时长的双因素交互作用均对微生物指标的测定有显著影响(P<0.05),其中时长的影响更大。基于加标试验和实际水样检测结果,建议生活饮用水水样的保存条件根据运输时长选择:若水样采集后4 h内检测,可于环境温度下保存;若12 h内检测,需于4~25℃条件下保存;若超过12 h检测,需于4~15℃冷藏保存水样,时长最长不超过24 h。Water sample collection and transportation are key steps that affect the objective and real level of microbial pollution in water.In this study,the total count of bacteria colonies was examined to investigate the effect of preservation temperature and preservation time of water samples on microbial indexes determination.Results showed that preservation temperature and time before examination,and the joint action of them significantly affected microbial determination(P<0.05),while the preservation time of water samples expressed the most obvious impact compared to other factors.Based on the determination results in this study,it was recommended that the preservation conditions of water samples should be selected according to the preservation time of samples:samples with preservation time less than 4 hours can be preserved at ambient temperature,less than 12 hours need to be preserved at 4~25℃,exceed 12 hours need to be preserved at 4~15℃with 24 hours as absolute maximum.
