摘　　要：目的探究HIST1H3B表达量变化对结直肠癌细胞功能的影响。方法使用RT-qPCR技术和Western blot分别检测HIST1H3B在SW1116细胞中的表达;利用Transwell实验检测HIST1H3B对细胞的迁移及侵袭能力的影响。结果通过RT-qPCR、Western Blot检测到SW1116过表达和沉默表达的细胞系构建成功。细胞功能实验表明HIST1H3B过表达促进细胞的迁移及侵袭功能,沉默HIST1H3B可抑制细胞迁移及侵袭功能。结论HIST1H3B上调表达可促进结直肠癌细胞的迁移和侵袭功能,可作为结直肠癌诊断和治疗的潜在生物标志物。Objective This study aimed to research the effect of HIST1H3 Boverexpression/knockdown on colorectal cancer cell function.Methods The expression levels of HIST1H3 Bin SW1116 cell were verified by Western blot and real-time quantitative PCR.The effect of HIST1H3 Bon colorectal cancer migration and invasion ability was studied by Transwell chamber migration and invasion assay.Results RT-qPCR and western blot analysis determined that we successfully constructed HIST1H3 Boverexpressed and silenced cell lines.After overexpression HIST1H3 B,cell function experiments show that the ability of migration and invasion of CRC cells was promoted.After silencing HIST1H3 B,the migration and invasion of CRC cells was inhibited.Conclusion Up-regulation of HIST1H3 Bcontributed to the migration and invasion of colorectal cancer cells.The results of immunohistochemical verify that HIST1H3 B is up-regulated in colorectal cancer.
