骡鸭大、小肥肝脂肪酸组分及脂代谢相关基因表达差异的对比分析

摘  要:为了研究骡鸭大、小肥肝的组织形态、脂肪酸组分以及脂代谢相关基因的表达差异,试验选取同批次且体重相近的300只公骡鸭在相同饲养管理条件下统一填饲19 d,根据屠宰时肝脏重与肝体比(肝脏重/体重)分为大肝组(HC组)和小肝组(LC组),分别制备HC组和LC组的肝脏组织切片,应用气相色谱质谱联用仪检测分析各组脂肪酸组分,实时荧光定量PCR方法检测SCD1、FADS2、FABP4、FASN、PDHB等脂代谢相关基因的相对表达量。结果表明:HC组个体体重、肝脏重、肝体比的平均值分别为5 828.57 g、893.57 g和15.37%,LC组个体分别为5 424.29 g、351.57 g和6.50%。骡鸭肥肝中在汇管区胆管周围有炎性细胞浸润,其中LC组有更多炎性细胞浸润。HC组饱和脂肪酸总含量极显著高于LC组(P<0.01),而LC组多不饱和脂肪酸含量极显著高于HC组(P<0.01),两组单不饱和脂肪酸含量差异不显著(P>0.05)。HC组和LC组单不饱和脂肪酸占比相似,分别为58.78%和59.33%;HC组饱和脂肪酸占比为36.25%,高于LC组的32.58%;HC组多不饱和脂肪酸占比为4.97%,低于LC组的8.09%。HC组中FASN、FADS2基因相对表达量极显著低于LC组(P<0.01),SCD1基因相对表达量显著低于LC组(P<0.05),FABP4基因相对表达量极显著高于LC组(P<0.01),PDHB基因相对表达量两组间差异不显著(P>0.05)。说明骡鸭大、小肥肝脂肪酸含量不同,小肥肝具有更多的不饱和脂肪酸,肝脏组织脂肪酸含量可能受其候选基因表达量的影响。In order to study the differences in tissue morphology, fatty acid composition and lipid metabolism-related gene expression in large and small duck fatty liver, 300 male mule ducks from the same batch with similar body weight were selected and overfed for 19 d under the same conditions, and then were divided into high capacity group(HC group) and low capacity group(LC group) according to liver weight and liver-to-body weight ratio(liver weight/body weight) at slaughter. The liver tissue sections of HC group and LC group were prepared, and the fatty acid components of each group were detected by gas chromatography-mass spectrometry. The expression of related genes(SCD1, FADS2, FABP4, FASN, and PDHB) was detected by fluorescence quantitative PCR. The results showed that the average body weight, liver weight and liver-to-body ratio in the HC group were 5 828.57 g, 893.57 g and 15.37%, respectively, and those in the LC group were 5 424.29 g, 351.57 g and 6.50%, respectively. In the fat liver of mule duck, there was inflammatory cell infiltration around the bile duct in the portal area, and there was more inflammatory cell infiltration in the LC group. The total contents of saturated fatty acids in the HC group were extremely significantly higher than that in the LC group(P<0.01), and the content of polyunsaturated fatty acids in the LC group was extremely significantly higher than that in the HC group(P<0.01), but there was no significant difference in the content of monounsaturated fatty acids between the two groups(P>0.05). The proportion of monounsaturated fatty acids in HC group and LC group was similar, which was 58.78% and 59.33%, respectively. The proportion of saturated fatty acids in the HC group was 36.25%, which was higher than that in the LC group(32.58%), but the proportion of polyunsaturated fatty acids in the HC group was 4.97%, which was lower than that in the LC group(8.09%).
