miR-338-3p调控MORC4促进宫颈癌细胞紫杉醇化疗敏感性

摘　　要：目的探究miR-338-3p通过调控MORC4表达对宫颈癌细胞紫杉醇(taxol,TAX)化疗敏感性的影响。方法实时荧光定量PCR(RT-qPCR)检测miR-338-3p的表达水平;MTT检测宫颈癌紫杉醇耐药细胞株(HeLa/TAX)细胞增殖活力;Transwell检测HeLa/taxol细胞侵袭能力;Annexin V-FITC/PI检测HeLa/TAX细胞凋亡水平;免疫印迹蛋白(Western blotting)检测HeLa/TAX细胞中MORC家族的CW型锌指(4Microrchidia4,MORC4)表达水平。结果miR-338-3p在宫颈癌细胞中异常低表达,且在TAX耐药细胞HeLa/TAX中表达较亲本HeLa细胞更低;过表达miR-338-3p可上调TAX促进HeLa/TAX细胞凋亡的作用和抑制细胞侵袭的作用;miR-338-3p靶向负调控MORC4表达;miR-338-3p可通过靶向下调MORC4表达,上调TAX对HeLa/TAX细胞增殖和侵袭的抑制作用,以及诱导细胞凋亡。结论过表达miR-338-3p可通过靶向下调MORC4表达,上调TAX对HeLa/TAX细胞凋亡的促进作用和对细胞侵袭能力的抑制作用,以此促进宫颈癌细胞对TAX化疗敏感性。Objective To investigate the effects of miR-338-3 p on paclitaxel(TAX)sensitivity of cervical cancer cells by regulating MORC4 expression.Methods Expression of miR-338-3 p was detected by Real-time fluorescence quantitative PCR(RT-qPCR).MTT detected the proliferation activity of HeLa/TAX cells.The invasion ability of HeLa/TAX cells was detected by Transwell.Annexin V-FITC/PI detected the apoptosis rate of HeLa/TAX cells.The MORC family CW-type zinc finger 4(Microrchidia4,MORC4)expression in HeLa/TAX cells was detected by Western blotting.Results miR-338-3 p was poorly expressed in cervical cancer cells,and was lower in TAX resistant HeLa/TAX cells than in parent HeLa cells.Overexpression of miR-338-3 p promoted the role of TAX in promoting HeLa/TAX cell apoptosis and inhibiting cell invasion.miR-338-3 p negatively regulated MORC4 expression.miR-338-3 p promoted the chemotherapy-sensitivity of HeLa to TAX through targeting to down-regulate the expression of MORC4.Conclusion Overexpression of miR-338-3 p promote the effects of TAX on promoting HeLa/TAX cell apoptosis and inhibiting cell invasion by down-regulating MORC4 expression,so as to promote the sensitivity of cervical cancer cells cells to TAX chemotherapy.
