摘 要:目的对槟榔多糖多酚的致畸作用进行探索性研究,为其风险评价、应用和人类健康提供依据。方法采用GB15193.14—2015《食品安全国家标准》中致畸试验方法对槟榔多糖多酚进行致畸试验研究。Wistar大鼠210只,雄、雌(1∶1)单笼交配,雌鼠阴栓阳性(或阴道涂片检出精子)日为妊娠0 d,随机分到各组。按照0、0.28、0.83和2.50 g/kg剂量经口灌胃染毒(孕期第6~15 d的器官形成期),于妊娠第20 d解剖孕鼠。称量、记录孕鼠体质量、仔鼠体质量及身长;检查胎鼠外观、骨骼、内脏及身体发育指标。结果 2.5 g/kg剂量组孕鼠体质量(12、15、18 d)、总增重、净增重及子宫重量(连胎重)、活胎百分率均低于对照组,窝平均吸收胎数及窝平均死胎数均高于对照组(均P<0.05)。胎鼠体质量及身长均低于对照组,且差异均有统计学意义(均P<0.05)。0.83 g/kg剂量组孕鼠体质量(15 d)低于对照组,且差异有统计学意义(P<0.05)。结论槟榔多糖多酚作为食用产品的主要原材料在摄入量低于0.28 g/kg时未见明显的胚胎毒性及致畸作用。Objective To explore the teratogenicity of areca nut polysaccharide polyphenols,provide evidence for its risk assessment,application and human health. Methods The teratogenic test of areca nut polysaccharide polyphenols was carried out according to GB 15193.14-2015 National Food Safety Standard. 210 Wistar rats,male and female(1 ∶ 1) mating in a single cage,were randomly divided into each group on the day of 0 day of pregnancy when the female rats were positive for the vaginal suppository(or the sperm was detected by vaginal smear). The pregnant rats were dissected on the 20 th day of gestation at doses of 0,0.28,0.83,2.50 g/kg by oral administration(organogenesis period from 6 th to 15 th day of gestation). The body weight of pregnant rats,and weight and body length of offspring were measured and recorded. The appearance,skeleton,viscera and body development indicators of fetal rats were detected. Results The body weight(12,15,18 d),total weight gain,net weight gain,uterine weight(even fetal weight),and live fetal percentage of pregnant rats in the 2.5 g/kg dose group were all lower than those in the control group,the mean number of foetus absorbed and foetus stillborn were higher than the control group(all P<0.05). The body weight and body length of the fetal rats were lower than those of the control group. The differences were statistically significant(all P<0.05). The weight of pregnant rats in the 0.83 g/kg dose group(15 d) was lower than that in the control group,and the differences were statistically significant(P<0.05). Conclusion As the main raw material of edible products,areca polysaccharide polyphenols shows no obvious embryonic toxicity and teratogenicity when the intake is lower than 0.28 g/kg.
