国产人工视网膜视觉假体植入兔眼的短期安全性和稳定性的研究

摘　　要：目的评估国产人工视网膜视觉假体植入兔眼后的短期安全性和稳定性。方法选用健康白毛黑眼兔10只,右眼为实验眼。予以全身麻醉后,植入眼行晶状体联合玻璃体切除,将人工视网膜视觉假体植入眼内视网膜上,并用视网膜钉固定。术中观察角膜是否透明、前房出血、眼内出血、视网膜脱离、人工视网膜视觉假体电极芯片的位置;分别于术后1、7 d,1、3、6个月观察角膜是否透明、前房出血与术后并发症发生情况(如眼内出血、视网膜脱离、眼内炎症情况),利用光学相干断层成像术、眼底照相及B超检查评估人工视网膜视觉假体电极芯片与视网膜的位置。6个月时处死动物摘除眼球行病理切片评估视网膜结构完整性。结果共10眼顺利完成人工视网膜视觉假体植入术;其中1眼术中出现眼内出血,并于1个月后吸收,人工视网膜视觉假体电极芯片位正。余9眼术后无明显眼内出血、未见视网膜脱离。6个月时病理切片显示视网膜变薄。结论国产人工视网膜视觉假体手术植入活兔短期观察安全可靠。Objective To evaluate the short-term safety and stability of implantation of the domestic-made retinal prosthesis in rabbits.Methods Ten white rabbits with black eyes were selected in the study,and the right eyes were used for experiments and the left ones were used for controls.After general anesthesia,the lensectomy and vitrectomy were performed,then retinal prosthesis was implanted onto the retina.Corneal transparency,intraocular hemorrhage and inflammation,retinal detachment and location of retinal prosthesis were observed during the operation,and 1 day,1 week,1 month,3 months and 6 months after operation.Fundus photography,optical coherence tomography(OCT)and ultrasonography were performed to evaluate the location of retinal prosthesis and retinal detachment.The animals were sacrificed 6 months after operation,and HE staining was used for pathological examination of the retinal structure.Results The implantation of retinal prosthesis was successfully completed in all animals.The intraocular hemorrhage occurred during operation in one eye,it was absorbed one month later,and the retinal prosthesis was in right position without retinal detachment.In other 9 eyes,there was no obvious intraocular hemorrhage and inflammation,the location of retinal prosthesis was in the primary place without retinal detachment and retinal hemorrhage.Retinal thinning was discovered in pathological examination.Conclusion The implantation of domestic-made retinal prosthesis in rabbits were proved to be safe and stable in the short-term observation.
