摘 要:目的通过生物信息学方法分析抗沉默功能蛋白1B(ASF1B)在肺腺癌组织中的转录水平表达及其临床意义。方法利用Oncomine、基因表达谱动态分析(GEPIA2)、UALCAN、Kaplan-Meier Plotter、Linked Omics数据库检索数据,比较ASF1B在常见肿瘤中的表达情况,并分析ASF1B在肺腺癌组织与正常肺组织中的表达水平,及其与临床分期和TP53突变的关系;分析ASF1B mRNA表达水平与细胞周期调控因子表达量的相关性;探索ASF1B mRNA表达与患者预后的相关性;最后检索ASF1B的共表达基因,将相关性最高的前50个基因生成在线热图。结果Oncomine数据库中共收集401项ASF1B mRNA在不同类型肿瘤组织与正常组织中表达比较的研究,其中47项ASF1B mRNA表达差异有统计学意义,有4项研究提示肺腺癌组织中ASF1B mRNA表达水平均高于正常肺组织(均P<0.05)。进一步分析GEPIA2和UALCAN数据库再次证实ASF1B的高表达,并且与患者的TNM临床分期和TP53突变均有关(均P<0.05)。ASF1B mRNA表达水平与大部分细胞周期蛋白和细胞周期蛋白依赖性激酶之间存在正相关表达关系。ASF1B高表达与患者的不良预后有关。ASF1B共表达基因分析结果分别显示了正、负相关性最高的前50个基因,初步探讨ASF1B参与肺腺癌进程的潜在分子机制。结论ASF1B在肺腺癌组织中高表达,且初步显示与肺腺癌的发生、发展及预后存在一定相关性,有望成为肺腺癌患者治疗的潜在靶点。Objective To analyze the mRNA expression of anti-silencing function 1B(ASF1B)in lung adenocarcinoma tissues and clinical significance based on bioinformatics.Methods Data were extracted from the databases Oncomine,GEPIA2,UALCAN,Kaplan-Meier Plotter and Linked Omics.The expression levels of ASF1B in lung adenocarcinoma and normal lung tissues were compared,its relationship with clinical stage,TP53 mutation,cell cycle regulators and the survival of patients was analyzed.The genes related to ASF1B expression was explored and the first 50 genes were generated as heat maps.Results A total of 401 different studies were collected in the Oncomine database,of which 47 showed that ASF1B was significantly expressed in cancer tissues,and 4 studies involved lung adenocarcinoma.The GEPIA2 and UALCAN databases confirmed the high expression of ASF1B in cancers,and it was significantly related to the patient's clinical stage and TP53 mutation.GEPIA2 database showed that ASF1B was correlated with cell cycle regulators Cyclins and CDKs.High expression of ASF1B was significantly associated with poor prognosis of patients.The heat map showed the top 50 ASF1B co-expression genes which were either positively or negatively correlated with ASF1B expression,and the potential molecular mechanism of ASF1B expression in the occurrence and development of lung adenocarcinoma was explored.Conclusion ASF1B is highly expressed in lung adenocarcinoma tissues,and it may be related to the occurrence,development and prognosis of lung adenocarcinoma.
