肝癌组织中miR-206与VEGFA表达的相关性及临床意义

摘　　要：目的研究肝癌组织中miR-206与血管内皮生长因子A(VEGFA)表达的相关性及临床意义。方法回顾性选择2017年12月至2020年1月期间在郴州市第一人民医院接受手术切除的110例肝癌患者,收集肝癌组织和距离癌组织2 cm以上的癌旁组织,检测miR-206、VEGFA的表达量;随访患者的总生存时间及无进展生存时间。分析肝癌组织中miR-206与VEGFA的相关性。在Targetscan网站进行miR-206靶向VEGFA的生物信息学分析。结果与癌旁组织比较,肝癌组织中miR-206的表达量降低(0.45±0.13 vs.0.79±0.15),VEGFA的表达量增加(0.93±0.19vs.0.42±0.09),差异均有统计学意义(P<0.05);TNMⅢ期、低未分化、有血管侵犯的肝癌组织中miR-206的表达量明显低于TNMⅠ-Ⅱ期、高中分化、无血管侵犯的肝癌组织,VEGFA的表达量明显高于TNMⅠ~Ⅱ期、高中分化、无血管侵犯的肝癌组织,差异均有统计学意义(P<0.05)。肝癌中miR-206高表达患者的无进展生存期、总生存期较miR-206低表达患者延长,肝癌中VEGFA高表达患者的无进展生存期、总生存期较VEGFA低表达患者缩短,差异有统计学意义(P<0.05)。相关性分析显示miR-206与VEGFA的表达水平呈负相关(r=-0.231,P<0.05)。生物信息学分析显示miR-206能够靶向结合VEGFA mRNA 3’UTR的第1526-1532碱基。结论肝癌中miR-206表达降低与VEGFA表达增加存在相关性,且与病理特征、远期预后有关。Objective To study the correlation of miR-206 and vascular endothelial growth factor A(VEGFA)expression in hepatocellular carcinoma and its clinical significance.Methods 110 patients with hepatocellular carcinoma who underwent surgical resection in Chenzhou First People’s Hospital from December 2017 to January 2020 were retrospectively selected to collect hepatocellular carcinoma tissues and adjacent tissues more than 2 cm away from cancer tissue,and the expression of miR-206 and VEGFA were determined,the total survival time and progression-free survival time of patients were followed up.The correlation between miR-206 and VEGFA in liver cancer tissues was analyzed.The bioinformatics analysis of miR-206 targeting VEGFA was carried out on Targetscan website.Results The expression of miR-206 in hepatocellular carcinoma tissues was significantly lower than that in adjacent tissues(0.45±0.13 vs.0.79±0.15),and the expression of VEGFA was significantly higher than that in adjacent tissues(0.93±0.19 vs.0.42±0.09),the differences were statistically significant(P<0.05);the expression of miR-206 in hepatocellular carcinoma tissues with TNMⅢstage,low undifferentiated stage and vascular invasion was significantly lower than that in hepatocellular carcinoma tissues with TNMⅠ-Ⅱstage,high or middle differentiation stage and non-vascular invasion,and the expression of VEGFA was significantly higher than that in hepatocellular carcinoma tissues TNMⅠ-Ⅱstage,high or middle differentiation stage and non-vascular invasion,the difference was statistically significant(P<0.05).Progression-free survival and total survival of patients with high expression of miR-206 were better than those with low expression of miR-206;progression-free survival and total survival of patients with high expression of VEGFA were worse than those with low expression of VEGFA,the differences were statistically significant(P<0.05).Correlation analysis showed that miR-206 was negatively correlated with the expression level of VEGFA(r=-0.231,P<0.0
