摘 要:目的:探讨6,7,4'-三羟基异黄酮(THIF)对人宫颈癌HeLa细胞免疫逃逸的影响及作用机制。方法:体外培养HeLa细胞,随机分为对照组、THIF组(5、15、30μmo/l L);MTT法检测THIF对HeLa细胞的增殖抑制作用及淋巴细胞增殖活力;LDH法检测自然杀伤(NK)细胞杀伤活力;Annexin V-FITC/PI双染法检测各组HeLa细胞凋亡情况;RT-qPCR和Western blot检测主要组织相容性复合体Ⅰ类链相关蛋白A/B(MICA/B)、可溶性NK细胞活化性受体(NKG2D)mRNA及蛋白表达。结果:与对照组相比,THIF各组HeLa细胞存活率依次降低(P<0.05),HeLa细胞凋亡率、淋巴细胞增殖活力、NK细胞杀伤活力、MICA、MICB、NKG2D mRNA及蛋白表达均升高(P<0.05),且呈剂量依赖性。结论:THIF可能通过上调MICA/B表达抑制宫颈癌细胞免疫逃逸,为THIF应用于宫颈癌临床治疗提供参考。Objective:To investigate effect of 6,7,4'-trihydroxyisoflavone(THIF)on immune escape of human cervical cancer HeLa cells and its mechanism.Methods:HeLa cells were cultured in vitro and randomly divided into control group and THIF groups(5,15,30μmo/l L);MTT method to detect inhibitory effect of THIF on HeLa cell proliferation and proliferation activity of lymphocytes.Cytotoxicity of natural killer(NK)cells was detected by LDH.Apoptosis of HeLa cells was detected by Annexin V-FITC/PI double staining;RT-qPCR and Western blot were used to detect mRNA and protein expressions of major histocompatibility complex classⅠchain-related protein A/B(MICA/B)and soluble NK cell group 2D(NKG2D).Results:Compared with control group,survival rates of HeLa cell in THIF groups were decreased in turn(P<0.05),HeLa cell apoptosis rate,lymphocyte proliferation activity,NK cell killing activity,MICA,MICB,NKG2D mRNA and protein expressions were increased in a dose-dependent manner(P<0.05).Conclusion:THIF might inhibit immune escape of cervical cancer cells by up-regulating expression of MICA/B,which provided reference for clinical treatment of THIF for cervical cancer.
