摘  要:目的应用超高效液相色谱-四极杆飞行时间串联质谱法(UHPLC-Q-TOF/MS)对长鞭红景天主要化学成分进行定性分析,并研究其体外抗氧化活性。方法采用Welch Ultimate UHPLC XB-C18(2.1mm×100 mm, 1.8μm)色谱柱,以0.1%甲酸水溶液(A)-乙腈(B)溶液为流动相,梯度洗脱,流速为:0.3 ml/min,柱温:40℃,电喷雾离子源,负离子模式下扫描采集数据;并测定其不同质量浓度的长鞭红景天提取物对DPPH、·OH自由基的清除能力。结果从长鞭红景天植物中共鉴定出36个化学成分,包括有机酚酸类、苯乙醇苷类、黄酮及萜类成分;当长鞭红景天提取物质量浓度达到30 mg/L时,其DPPH清除率为91.3%,而当其质量浓度为1 mg/ml时,其羟基自由基清除率为81.8%。结论该方法高效、准确、灵敏,可为长鞭红景天的质量控制提供依据,其抗氧化活性研究可为后续长鞭红景天的药效及药效物质研究提供基础。Objective UHPLC-Q-TOF/MS was used to qualitatively analyze the chemical components of Rhodiola fastigiata and its in vitro antioxidant activity. Methods The separation was performed on Welch Ultimate UHPLC XB-C18(2.1 mm×100 mm, 1.8 μm) column with gradient elution of 0.1% formic acid(A)-acetonitrile(B), the flow rate was 0.3 mL/min, The column temperature was set at 40 ℃, The MS analysis was based on information associated mode(IDA), and negative ion was collected;And evaluating the scavenging ability of Rhodiola fastigiata extracts with different concentrations on DPPH and ·OH. Results 36 chemical constituents were identified from Rhodiola fastigiata,including organic phenolic acids, phenethanol glycosides, flavonoids and terpenes;the concentration of Rhodiola fastigiata extract reaches 30 mg/L, its DPPH clearance rate was 91.3%, at the concentration of 1 mg/ml, its ·OH clearance rate was 81.8%. Conclusion This method is efficient, accurate and sensitive, and can provide technical support for the quality control of Rhodiola fastigiata, and its antioxidant activity can provide experimental evidence for the subsequent elucidation of the material basis of the efficacy of Rhodiola fastigiata.
