摘 要:以小球藻为原料,通过单因素实验探究硫酸铵质量分数、乙醇体积分数、超声功率、料液比和提取时间对多糖得率的影响。在此基础上,采用遗传算法优化超声辅助双水相提取小球藻多糖工艺,并对比不同提取方式对小球藻多糖得率的影响。结果表明超声辅助双水相提取小球藻多糖最优的工艺参数为:硫酸铵质量分数21%、乙醇体积分数22%、超声功率406W、料液比1∶22g/mL和提取时间30min。在此条件下,所得小球藻多糖得率为(6.31±0.02)%。实验值和理论值的相对误差为2.77%。表明遗传算法优化小球藻多糖提取工艺参数是可行的。与其他提取方式对比,超声辅助双水相提取是一种绿色高效的提取方式。The effect of ammonium sulfate mass fraction,ethanol concentration,ultrasonic power,solid-to-liquid ratio and extraction time on the yield of polysaccharide from Chlorella was investigated by single factor experiment.On this basis,genetic algorithm was used to optimize the ultrasonic assisted aqueous two-phase extraction process of Chlorella polysaccharide,and the effect of different extraction methods on the yield of Chlorella polysaccharide was compared. The results showed that the optimum extraction parameters to achieve the highest polysaccharide yield(6.31±0.02)% from Chlorella by ultrasonic-assisted aqueous two-phase extraction were obtained under an ammonium sulfate mass fraction of 21%,ethanol concentration of 22%,ultrasonic power of 406 W,solid-toliquid ratio of 1∶22 g/mL and extraction time of 30 min. The relative error between the experimental value and the theoretical value was 2.77%. The results showed that it was feasible to optimize the extraction parameters of Chlorella polysaccharide by genetic algorithm. Compared with other extraction methods,ultrasonic assisted aqueous two-phase extraction is a green and efficient extraction method.
