摘 要:本试验旨在研究黄曲霉毒素M_(1)(AFM_(1))与赭曲霉毒素A(OTA)对小鼠肠道内微生物菌群结构的影响。试验选用健康的48只4周龄ICR小鼠,随机分成4组,每组12只,对照组灌胃1%二甲基亚砜(DMSO)溶液,AFM_(1)组灌胃3.5 mg/kg BW AFM_(1),OTA组灌胃3.5 mg/kg BW OTA,AFM_(1)+OTA组灌胃3.5 mg/kg BW AFM_(1)+3.5 mg/kg BW OTA,每天灌胃1次,连续灌胃35 d。灌胃结束后,采用16S rRNA测序对小鼠结肠微生物多样性进行分析。结果表明:与对照组相比,AFM_(1)与OTA处理后,不论单独或者混合处理都会导致厚壁菌门、乳杆菌科、乳酸菌属、益生菌拟普雷沃菌属相对丰度显著降低(P<0.05);拟杆菌门、变形菌门、瘤胃球菌科、肠杆菌科、梭菌属相对丰度显著增加(P<0.05)。综合试验结果表明,AFM_(1)、OTA单独及联合处理均改变了小鼠肠道内微生物菌群结构,损伤了肠道微生物屏障功能。This study was aimed to investigate the effects of aflatoxin M_(1)(AFM_(1))and ochratoxin A(OTA)on the structure of intestinal microflora of mice.A total of 48 healthy 4-week-old ICR mice were selected and randomly divided into 4 groups with 12 mice in each group.The control group was gavaged with 1% dimethyl sulfoxide(DMSO)solution,the AFM_(1) group was gavaged with 3.5 mg/kg BW AFM_(1),OTA group was gavaged with 3.5 mg/kg BW OTA,and the AFM_(1)+OTA group was gavaged with 3.5 mg/kg BW AFM_(1)+3.5 mg/kg BW once a day for 35 days.After intragastric administration,microbial diversity in the colon of mice was analyzed by 16 S rRNA sequencing.The results showed that after AFM_(1) and OTA treatment,compared with control group,both single and mixed treatments resulted in decreased relative abundance of Firmicutes,Lactobacillaceae,Lactobacillus and Alloprevotella(P < 0.05),and increased relative abundance of Bacteroidetes and Proteobacteria,Ruminococcaceae and Enterobacteriaceae,Clostridium_Sensu_Stricto_1(P<0.05).These results indicate that both AFM_(1) and OTA treatments change the structure of intestinal microflora and impair the function of intestinal microbial barrier in mice.
