摘  要:本研究旨在评估热应激奶牛乳腺组织中差异表达miRNA及其在乳蛋白合成调控中的潜在作用。选取4头泌乳日龄、体重、产奶量相近的经产健康荷斯坦奶牛作为试验动物。采用2×2交叉设计,试验分2期,每期包含预试期和试验期各9 d,2期之间间隔30 d。试验在大家畜人工环境控制舱内进行。预试期为热中性环境,自由采食。试验期奶牛随机分为2组,分别为热应激组(HS组,n=4)和配对限饲组(PFTN组,n=4)。结果表明:1)16个miRNA在HS组和PFTN组奶牛乳腺组织中差异表达(Padj<0.05)。2)差异表达的miRNA中bta-miR-451可能参与了能量代谢的调控,bta-miR-184、bta-miR-218和bta-miR-493可能参与了细胞增殖的调控,bta-miR-132可能参与了炎症反应的调控,而bta-miR-382与乳腺组织中mRNA表达调控可能存在协同作用。由此可见,热应激奶牛乳腺组织中miRNA可能参与了能量代谢、细胞增殖和炎症反应的调控,为进一步验证miRNA在热应激诱导的乳蛋白合成减少过程中的作用提供了科学依据。The objective of the present study was to evaluate the differently expressed miRNA in dairy cows’ mammary tissue and its potential roles in regulation of milk protein synthesis.Four healthy Holstein cows with similar lactation age,body weight and milk yield were selected as experimental animals.A 2 ×2 crossover design was used,the experiment included two periods,each period included pre-test period and test period for 9 days,respectively,and the interval between two periods was 30 days.The experiment was carried out in the artificial environment control cabin.The pre-test period was a hot neutral environment and free feeding.Cows in test period were randomly divided into two groups:heat stress group(HS group,n = 4)and pair-feeding restriction group(PFTN group,n = 4).The results showed as follows:1)sixteen differently expressed miRNA were identified in dairy cows’ mammary tissue between HS group and PFTN group(Padj<0.05).2)In differently expressed miRNA,bta-miR-451 might involve in the regulation of energy metabolism,bta-miR-184,bta-miR-218 and bta-miR-493 might involve in the regulation of cell proliferation,bta-miR-132 might involve in the regulation of inflammation response,while bta-miR-382 might have synergistic effect with the regulation in mRNA expression in mammary tissue.In conclusion,miRNA in mammary tissue may involve in the regulation of energy metabolism,cell proliferation and inflammatory response,this study lays a theoretical and scientific foundation for further studies to confirm the regulatory effect of miRNA on milk protein synthesis under heat stress.
