摘 要:目的建立乳粉中总肌醇的盐酸水解提取-硅烷化衍生-气相色谱测定法。方法乳粉通过盐酸水解,加无水乙醇提取后氮吹浓缩,与硅烷化试剂衍生,正己烷提取,经气相色谱分离,外标法定量。分别用盐酸水解提取法和乙醇溶解直接提取法提取样品并测定,比较两方法的提取率。结果本方法在1.0μg/ml~50.0μg/ml浓度范围内,线性关系良好,相关系数为0.9997,方法检出限为1.0 mg/100 g,平均回收率为88.83%~95.94%,相对标准偏差为2.14%~4.12%。盐酸水解提取法提取出乳粉中的结合态肌醇,测定结果为总肌醇含量,高于乙醇溶解直接提取法。结论本方法操作简单、快速、准确,适用于乳粉中总肌醇含量的测定。Objective A method for the determination of total inositol in milk powder by hydrochloric acid hydrolysis extraction-silanization derivation-gas chromatography was established.Methods The milk powder was hydrolyzed with hydrochloric acid and extracted with anhydrous alcohol,then concentrated by nitrogen blowing,derived by silanizing reagent.The derived product was extracted with n-hexane,and separated and detection by gas chromatography and quantified by external standard method.Hydrolytic extraction with hydrochloric acid and direct extraction with ethanol dissolution were used to extract and determine the extraction rates of the two methods.Results In the range of 1.0μg/ml-50.0μg/ml,the linear correlation coefficient was 0.9997,the detection limit was 1.0 mg/100 g,the average recoveries were 88.83%-95.94%and the relative standard deviation were 2.14%-4.12%.The results of hydrochloric acid hydrolysis extraction method can extract the bound inositol from milk powder,which is higher than that of ethanol dissolution extraction method.Conclusion The method is simple,rapid,accurate and suitable for quantitative determination of inositol in milk powder.
