摘 要:目的:分别检测常温干燥区、常温潮湿区和4℃冰箱中储存饵块真菌菌落总数,分子生物学方法进行鉴定,为选择饵块储存方式提供参考依据。方法:取25 cm;饵块置于常温干燥区、常温潮湿区和4℃冰箱中储存,48、72、96、120 h分别计数菌落总数。挑取菌体接种于YPD培养基中增菌提取DNA,PCR扩增ITS序列,测序后NCBI中进行比对。结果:常温潮湿区饵块污染真菌菌落总数最大,常温干燥区次之,4℃冰箱最少。分离到23株菌,分属于2个属,酵母菌属Saccharomyces和青霉菌属Penicillium,4个种,酿酒酵母Saccharomyces cerevisiae、异常毕赤酵母Pichia anomala、粘红酵母Rhodotorula mucilaginosa、青霉菌Penicillium crustosum。结论:常温潮湿区储存饵块湿润度和柔软度最好,但最易被青霉菌和酵母菌污染,干燥区储存饵块不易受酵母菌污染。Objective The total number of fungal colonies stored were detected in the dry area,wet area and 4℃refrigerator,then the species were identified by molecular biology method.The appropriate storage mode of Er Kuai was provided base on the research.Methods The Er Kuai was stored under three environments and the number of bacteria was observed at different time.After the separation and culture,DNA extraction,PCR amplification and sequencing were carried out,and all ITS sequences obtained were analyzed and compared in NCBI.Results The highest total number of fungus was store in the wet area,followed by the dry area,and the lowest in the refrigerator at 4℃.23 strains were isolated,which were divided into two genera,Saccharomyces and Penicillium,with 4 species,Saccharomyces cerevisiae,Pichia anomala,Rhodotorula mucilaginosa and Penicillium crustosum.Conclusion The wetness and softness of Er Kuai stored in wet area were the best,but they were most easily contaminated by penicillium and Saccharomyces,while those stored in dry area were not suitable for Saccharomyces.
