摘 要:目的观察补肾调经方(Bu Shen Tiao Jing Decoction, BSTJD)对人子宫内膜微血管内皮细胞(Human Endometrial Microvascular Endothelial Cells, HEMECs)中雌激素受体α(ERα)和wnt/β-catenin信号通路的影响,阐明BSTJD促进血管生成改善子宫内膜容受性的作用机制。方法将动情周期正常的昆明系雌性小鼠随机分为正常组、模型组和补肾组,每组40只小鼠。建立控制性超促排卵(Controlled ovarian hyperstimulation, COH)的小鼠模型,造模第1天补肾组灌服BSTJD 0.4ml/30g(浓度1.5g/ml),正常组和模型组用生理盐水代替灌胃药物,连续11天。孕4天取子宫组织,免疫荧光观察子宫内膜血供情况;孕8天观察小鼠胚胎种植数;培养HEMECs并给予BSTJD预孵育,用Western blot、实时荧光定量PCR(qRT-PCR)和免疫荧光方法来检测β-catenin和ERα的表达变化。结果 (1)BSTJD可提高模型组小鼠妊娠数、胚胎种植数,丰富子宫内膜血供。(2) BSTJD可促进模型组小鼠子宫中ERα的表达,与上调β-catenin的表达密切相关,进而提高子宫内膜容受性。结论 BSTJD促进血管生成改善子宫内膜容受性,与上调β-catenin的表达和诱导ERα表达有关。Objective To observe the effect of Bu Shen Tiao Jing Decoction(BSTJD) on estrogen receptor α(ERα) and wnt/β-catenin signaling pathway in human endometrial microvascular endothelial cells(HEMECs), and clarify the mechanism of BSTJD promoting angiogenesis and improving endometrial receptivity.Methods Female Kunming mice, whose estrus cycle is normal, were randomly divided into 3 groups: normal group, model group and Bu Shen group, with 40 mice in each group. Except for the normal group, mice in the other groups were established a mouse model of controlled ovarian hyperstimulation(COH). On the first day of model preparation, Bu Shen group is given 0.4 ml/30 g of BSTJD with 1.5 g/ml, and the normal group and the model group used normal saline instead of gavage for 11 consecutive days. We take the uterine tissue immunofluorescence to observe the blood supply of the endometrium on 4 days of pregnance;we observe the numbers of embryo implantation in mice on 8 days of pregnance. We culture HEMECs and pre-incubate with BSTJD, detect changes in the expressions of β-catenin and ERα by Western blot, quantitative real-time PCR(qRT-PCR) and immunofluorescence methods.Results(1)BSTJD can increase the numbers of pregnancy and embryo implantation in the model group, and enrich the blood supply of the endometrium.(2)BSTJD can promote the expression of ERα in the uterus of mice in the model group, which is closely related to the up-regulation of β-catenin expression, thereby promoting endometrial angiogenesis.Conclusion BSTJD induces ERα and β-catenin expressopm to promote angiogenesis, which improves endometrial receptivity.
