摘  要:锌指蛋白基因在植物生长发育过程中起着关键作用,尤其是在植物逆境生长中。为探究马铃薯锌指蛋白基因StZFP593在逆境胁迫中的表达特性,本研究以‘青薯9号’为材料,利用PCR扩增该基因,利用RT-qPCR方法分析马铃薯在晚疫病病菌诱导、低温以及钠盐处理下StZFP593在其各组织中的表达特性。结果表明:StZFP593全长661 bp,其中CDS序列长363 bp,编码120个氨基酸,不稳定指数(Ⅱ)计算为52.34>40,为不稳定蛋白。在晚疫病病菌诱导下,StZFP593在叶片中显著上调表达;在4℃处理下,StZFP593在根、茎、叶中的表达量均在72 h达到最高,在2℃处理下,该基因在根、茎中表达量分别在72、96 h达到最高,而在叶中均下调表达;在100 mmol/L钠盐处理5 d时,该基因在根、叶中的表达量显著上调表达,在茎中无显著差异表达,在150 mmol/L钠盐处理10 d时,该基因在根、叶中的表达量最高,而在茎中仍无显著差异表达。由此说明StZFP593在各组织中的表达受不同逆境胁迫诱导,参与了马铃薯在逆境胁迫的防御过程并发挥重要的作用。本研究可为后续全面深入研究马铃薯锌指蛋白基因的功能提供一定的理论依据。Zinc finger protein genes play a key role in the growth and development of plants,especially in the growth of plants under adversity.In order to explore the expression characteristics of the potato zinc finger protein gene StZFP593 under adversity stress,this experiment used’Qingshu 9’as the material and used PCR to amplify the gene.The RT-qPCR method was used to analyze the expression characteristics of StZFP593 in various tissues of potato under the induction of Phytophthora infestans,low temperature and sodium salt treatment.The result showed that the full length of StZFP593 was 661 bp,and the CDS sequence was 363 bp and encoded 120 amino acids.The instability index(Ⅱ)was calculated as 52.34>40,which is an unstable protein.Under the induction of Phytophthora infestans,the expression of StZFP593 was significantly up-regulated in leaves.Under 4℃treatment,the expression level of StZFP593 in roots,stems,and leaves reached the highest at 72 h.Under 2℃treatment,the expression of this gene in roots and stems reached the highest at 72 h and 96 h,respectively,and the expression was down-regulated in the leaves.When treated with a sodium concentration of 100 mmol/L for 5 days,the expression of this gene in roots and leaves was significantly up-regulated,but there was no significant difference in expression in the stem.When treated with a sodium concentration of 150 mmol/L for 10 days,the expression level of this gene is the highest in roots and leaves,but there is still no significant differential expression in stems.These results indicate that the expression of StZFP593 in various tissues is induced by different adversity stresses,and it participates in the defense process of potato under adversity stress and plays an important role.This study can provide some theoretical basis for the follow-up comprehensive and in-depth study of the function of the potato zinc finger protein gene.
